Before a tissue can be observe with a light
microscope, it must be made into a section. The
routine histological preparation for light microscope

examination is a paraffin section stained with


haematoxylin and eosin (abbreviate HE ).
This is a picture of muscle cells,
HE staining. The nucleus of the
cell is blue which stained
by haematoxylin(a basic dye ).
So the nucleus is said to
be basophilic.

The cytoplasm of the cell is


pink which stained by
eosin(an acidic dye ). So the
cytoplasm is termed acidophilic.

A cell includes three parts:
Cell membrane or plasma membrane
Cytoplasm
Nucleus

Smooth muscle cells are fusiform,. Each cell has a single , ellipsoid
nucleus located in the center of the broadest part of the cell. HE

Mitochondria can be seen with the light microscope in specially


stained preparations. They are so called because they appear either
as granules or as rods. .

In light microscopic preparations suitably treated with silver salts the


Golgi complex can be seen as a small structure of irregular shape,
usually present near the nucleus. The Golgi complex is the place where
the secretory products are packaged within membrane-bound vesicles.
