DENATURATION
PROCESS
•  the first regular cycling event and
consists of heating the reaction to
94–98 °C for 20–30 seconds
•  causes DNA melting of the DNA
template by disrupting the
hydrogen bonds between
complementary bases, yielding
single strands of DNA.
Annealing the primers
• Lowering the temperature to 55°C
causes the primers to bind to the DNA – A
process known as hybridization or
annealing
• The resulting bonds are stable only in the
primer and the DNA segments are
complementary, if the base pairs of the
primers and DNA segment match

• The polymerases then begin to
attach additional complementary
nucleotides, thus strengthening the
bonding between the primers and
the DNA
