The current technology leads to the alternative energy sources as replacement
for non-renewable energy. One of them is the of biodiesel manufacture as a subtitution of diesel fuel. Microalgae are a viable source of renewable energy to be developed. Botryococcus braunii and Nannochloropsis sp. are microalgae that currently being tested for its ability as diesel fuel source because of its high lipid content. Mutagenesis were done to improve the productivity of lipids, which used UV B rays as mutagen. The first stage was to do microalgae Botryococcus braunii and Nannochloropsis sp. cultivation for 14 days and nutrients adding once every 12 hours. The second stage was to determine the effect of UV-B ray mutagen on the growth of microalgae in different medium pH. The third stage was lipid extraction. Lipid extraction process steps include of centrifugation, drying in an oven at 60 C for 2 hours, soxhlet extraction using n-hexane solvent, separation of lipids from the solvent with a rotary vacuum evaporator. Furthermore GCMS analysis was used to determine the content of the compound of microalgae oil, DNA sequencing to determine the sequence of DNA bases her.
