Anti-Reverse Cap Analog

Description

n order to carry out large scale synthesis of RNA transcripts by way of in vitro transcription, adjustment of the RNA capping element is suggested. To help researchers accomplish this, Allele offers ARCA (Anti-Reverse Cap Analog), which is a modified cap analog in which the 3 OH group is replaced with OCH3. This re-figuration allows for RNA polymerase to only initiate transcription with the remaining hydroxyl group, and forces ARCA incorporation in the forward orientation. This outcome leads to the transcripts synthesized with ARCA at the 5 end to become translatable causing a stimulatory effect on translation. This feature is not found in standard cap analogs.
