CHAPTER 2 : CELL STRUCTURE AND CELL ORGANISATION

EXPERIMENT 1 ACTIVITY 2.1 : PREPARING AND EXAMINING SLIDES OF PLANT CELLS AIM : To prepare and study a slide of plant cells (epidermal cell of onion) PROBLEM STATEMENT : (No need for this experiment) HYPOTHESIS : (No need for this experiment) VARIABLES :(No need for this experiment) MATERIAL : An onion bulb, iodine solution and distilled water, APPARATUS : Light microscope, glass slides, cover slips, a scalpel, a pair of forceps, a glass dropper, a mounting needle and filter paper. PROCEDURE : 1. A scale of leaf is obtained from an onion bulb. 2. Using a pair of forceps, the transparent of epidermis is peeled off from the inner surface of onion scale leaf. 3. A drop of water is dropped into the middle of the slide and the epidermis is placed on the top of water. 4. With the help of a mounting needle, the specimen is covered with a cover slip at a 45 angle. 5. A drop of iodine solution is added onto one side of the cover slip. A filter paper is placed at the opposite end of the cover slip to allow the iodine solution to spread through the epidermis. 6. A piece of filter paper is used to absorb the excess of iodine solution. 7. The slide is examined under a light microscope using the low power objective lens and then the high power objective lens. 8. The epidermis is draw and the cell structures which can be observed is labeled. The magnification used is recorded. OBSERVATION : (draw the epidermis cell of onion and label. Write down the magnification power of the objective lens. 4x, 10 x or 40x)

ACTIVITY 2.2 : PREPARING AND EXAMINING SLIDES OF ANIMAL CELLS AIM : To prepare and study a slide of animal cells (human cheek cells) PROBLEM STATEMENT : (No need for this experiment) HYPOTHESIS : (No need for this experiment) VARIABLES :(No need for this experiment) MATERIAL : Methylene blue solution , iodine solution and distilled water, APPARATUS : Light microscope, glass slides, cover slips, a scalpel, a pair of forceps, a glass dropper, a mounting needle, filter paper and toothpicks. PROCEDURE : 1. The inside of the cheek is scraped with a toothpick. 2. The cheek extract is placed on the slide. A drop of water is dripped on it. 3. With the help of a long needle, a cover slide is gently lowered onto the specimen. Make sure there are no air bubbles trapped under the cover slip. 4. A drop of methylene blue solution is placed at one end of the cover slip and a strip of filter paper is placed at the other end of the cover slip. The filter paper will draw the solution across the specimen and colour it. This technique is called the irrigation technique. 5. The slide is then observed under the light microscope using the low-powered lens, followed by the highpowered lens. OBSERVATION : (draw the human cheek cell and label. Write down the magnification power of the objective lens. 4x, 10 x or 40x)

DISCUSSION : 1. Human cheek cells do not have fixed shape. 2. The colour of the cytoplasm and the nucleus that are stained with the methylene blue solution is .. 3. Yes/ no. I can/cannot see the dark blue spot on the cheek cells. 4. The cell structure that can be observed under the light microscope is . 5.
