Potential of phytase producing Bacillus licheniformis strain PB13 in plant growth promotion under greenhouse conditions

Abstract
In the present study phosphate solubilizing bacterial isolate from rhizospheric soil sample collected from Badrinath region of Uttrakhand, India, was screened for its plant growth potential. Preliminary identification of bacterial isolate as Bacillus licheniformis strain PB13, was made on the basis of morphological and biochemical characters and confirmed by partial 16S-rRNA gene sequencing. It was found to be solubilizing both forms of phosphorus i.e., organic and inorganic. The isolate was screened for its various plant growth promoting activities like production of ammonia, siderophore, auxin, phytase, HCN, and phosphorus solubilization. The isolate was capable of solubilizing phosphate, produced indole-3-acetic acid, ammonia in culture media and siderophore production on plates. On the basis of above criteria, green house studies were conducted with Brassica juncea to evaluate the potential of the isolate to promote plant growth. Effect of seed bacterization on shoot length, root length, fresh weight, dry weight and Phosphate (P) uptake by plant were studied. The isolates were found to be efficient in stimulating shoot length (40%) and fresh weight (30%) of mustard seedlings grown under greenhouse conditions. Significantly higher level of phosphate accumulation of 7.48 mg/g dry weight was noticed in seedlings raised from seeds bacterized with isolate PB13. From the results obtained Bacillus licheniformis strain PB13 appears to be a potential candidate as bio-inoculant. Key words: Phytase, Bacillus licheniformis, Plant growth promotion, Siderophore
