Anti-inflammatory effect of ethanol extract of sapu jagad (Isotoma longiflora (L) Presl.) leaves to leukocyte and neutrophil count in male mice (Mus musculus)

Phytochemical screening of ethanol extract ofsapu jagad (Isotoma longiflora (L) Presl.) leaves to revealed the presence of flavonoids, phenols, terpenoids and steroids. This compound suggested that sapu jagad possesses potent anti-inflammatory activity. The study aimed to investigate the effect of ethanol extract of sapu jagad leaves to leukocyte and neutrophil count in male mice which carrageenin-induced. Samples were male mice with 30-50 g bodyweight which were divided into five treatment groups (standard (aquadest 1 ml), negative control (carrageenin-induced mice and aquadest), and groups P1,P2 and P3 respectively to get the ethanol extract of sapu jagd leaves dose of 2, 6 and 10 mg/g body weight and carrageenininduced. Carrageenin 1%, 0.05 ml was used to induce inflammation, thirty minute after treatment. Six hour after the injection of carrageenin, mice were anaesthetized with ether. Blood samples were collected separately from each animal by cardiac puncture. Leukocytes counted by manual methods using counting chamber and neutrophils counted are analyzed on blood smears. The results showed leukocyte count statistic showed no significant difference (p>0.05) between standard groups with negative control groups and between negative control groups with groups P1, P2 and P3. Neutrophil count statistic showed significant difference (p<0.05) between standard groups with negative control groups and between negative control groups with groups P1, P2 and P3. In conclusion, ethanol extract of sapu jagad leaves could decrease leukocyte and neutrophil count. ABSTRAK Efek Antiinflamasi Ekstrak Etanol Daun Sapu Jagad (Isotoma longiflora (L) Presl.) Terhadap Jumlah Leukosit dan Jumlah Neutrofil pada Mencit Jantan (Mus musculus) Oleh Dini Mayrisdayani

Skrining fitokimia ekstrak etanol daun sapu jagad (Isotoma longiflora (L) Presl.) menunjukkan tumbuhan ini mengandung senyawa-senyawa kimia berupa flavanoid, fenol, terpenoid dan steroid. Senyawa yang terkandung dalam senyawa ini diduga memiliki efek antiinflamasi. Penelitian ini bertujuan untuk mengetahui efek antiinflamasi ekstrak etanol daun sapu jagad terhadap jumlah leukosit dan jumlah neutrofil pada mencit jantan. Sampel penelitian adalah mencit jantan dengan berat 30-50 g yang dibagi menjadi 5 kelompok dengan perlakuan berbeda (standar (aquades 1 ml), kontrol negatif (mencit yang diinduksi karagenin dan diberi aquades) dan kelompok P1, P2 dan P3 yang diberikan ekstrak etanol daun sapu jagad 2 mg/gBB, 6 mg/gBB dan 10 mg/gBB serta diinduksi karagenin. Karagenin 1%, sebanyak 0,05 ml digunakan sebagai penginduksi inflamasi, 30 menit setelah diberi perlakuan. Enam jam setelah penyuntikkan karagenan, mencit dianastesi dengan menggunakan eter. Pengambilan darah dilakukan dengan punksi jantung. Jumlah leukosit dihitung secara manual dengan menggunakan kamar hitung dan jumlah neutrofil dengan hapusan darah tepi. Penelitian ini menunjukkan jumlah leukosit tidak menunjukkan perbedaan bermakna (p>0,05) antara kelompok standar dan kelompok kontrol negatif dan antara kelompok kontrol negative dengan kelompok P1, P2 dan P3. Kesimpulannya adalah ekstrak etanol sapu jagad dapat menurunkan jumlah leukosit dan neutrofil.
