A look inside
With fluorescence flow cytometry
individual blood cells stained by
fluorochromes are illuminated by a
semi-conductor laser beam, enab-
ling a sophisticated analysis based
on RNA/DNA content, cell size, and
inner cell complexity. Looking into
the cell and judging the metabolic
activities in respect to cell replica-
tion and protein synthesis offers
much advanced possibilities for the
determination of the cell type and
maturation stage.
The collection and fast processing
of three optical signals obtained
from each individual cell are the
basis for the cell identification.
When it comes to white blood cell
differentiation, analysing the cell
interior properties rather than cell
size leads to reliable classification
also in older samples and clearly
reduces interferences.
Using sysmex unique adaptive
cluster analysis system (acas)
instead of fixed discriminators re-
sults in correct cell clustering and
thus WBC differential counts also in
highly pathologic samples, accord-
ingly reducing the false positive
rate. The results are illustrated in
informative scattergrams.
In the optical unit of xt-1800i
a semi-conductor laser, the flow cell,
optical focussing systems and detectors
are combined in a sophisticated system.
The forward scattered light signal
(cell size information), the side scattered
light signal (internal cell structure)
and the side fluorescence light signal
(RNA/DNA information) of each cell are
analysed as it passes through the flow
cell.

The various WBC populations and
immature precursor cells show a spe-
cific reaction with the fluorescent dye.

The xt information
processing unit (IPU)
The main unit and the complete
operation of the xt-1800i is controlled
by the operator via the information
processing unit with interactive
menus. Intuitively understandable
icons are used for the graphical user
interface of Windows.
In the IPU data base, all analysis re-
sults and patient data including the
complete set of scattergrams and
histograms of up to 10,000 samples
are stored.
Back-up facilities on CD-RW offer
principally unlimited storage capa-
city. Quality control programs allow
unattended analysis of control blood
samples in closed auto-sampler mode
or in manual mode.
The multi-tasking computer system
permits real time bidirectional com-
munication with the Host computer
to request patient profiles for
discrete testing with patient demo-
graphical information and transmits
analysis results immediately after
analysis is completed. Even if the
system is operated offline and with-
out bar code system, selective sample
analysis is maintained by using the
work list function.
Flexibility
The sysmex xt-1800i is available in
multiple configurations. This offers
high flexibility and satisfies even the
needs of the most demanding hae-
matology laboratory.
A 50-tube auto-sampler can be
mounted on the main unit of the
analyser offering walkaway auto-
mation. This sampler system enables
continuous feeding of samples with
a throughput rate of 80 samples per
hour. A STAT function allows imme-
diate analysis of emergency samples.
If desired, it is possible to install a
manual cap piercer system instead
of the sampler.
No matter whether a manual or an
automated bar code reader is used,
adequate positive sample identifica-
tion gives confidence.
The complete main unit operation and all data management functions are performed on the
information processing unit (IPU).
The optional sampler unit of xt-1800i
ensures efficient operation and through-
put of 80 samples per hour, independent
of the parameter profiles analysed.
