RESUMEN
El estudio de la taxonoma y filogenia de hongos, se ha basado en diversas tcnicas desde la
morfolgica hasta las moleculares. Debido a la propagacin sexual y asexual de los hongos
se gener un sistema dual en su nomenclatura bajo el enfoque morfolgico, el cual tiene
grandes limitaciones, sin embargo con el desarrollo de tcnicas de biologa molecular, y
particularmente con la estandarizacin para su aplicacin prctica y rpida, se ha podido
superar en parte esta limitacin, as mismo la aplicacin de estas metodologas permiten un
sistema de clasificacin que toma en cuenta la historia evolutiva de los hongos. Las
metodologas basadas en PCR han empleado diversas tcnicas tales como el RFLP, RAPD,
AFLP y DAF para la identificacin de hongos, y recientemente un enfoque integral (polifsico)
ha revolucionado la sistemtica de hongos, incluso se ha propuesto de manera formal una
nueva clasificacin en los taxa superiores del reino Fungi. La iniciativa del cdigo de barras
del ADN de la vida, propone una alternativa novedosa para la exploracin de la diversidad
biolgica, adems permite utilizar secuencias de ADN estandarizados para su aplicacin en
la identificacin a nivel de especies de los hongos de manera rpida, precisa y universal, con
el potencial de ser automatizada.
1.INTRODUCCIN
La posibilidad de una rpida identificacin de la diversidad biolgica o de un organismo a
nivel de especie, ha sido siempre deseable pero rara vez ha sido posible debido a la falta de
especialistas as como de las herramientas adecuadas para su correcta aplicacin (Chase y
Fay, 2009). Con el fin de desarrollar nuevas alternativas de identificacin se han propuesto
diversos mtodos moleculares que permiten no solo identificar a nivel de especie sino
tambin trazar la historia evolutiva de los individuos (Guarro y col.,1999). La estandarizacin
de estas herramientas para su aplicacin rpida y precisa en la identificacin de especies se
conoce como el cdigo de barras de la vida (Stockinger y col., 2010).
Para la implementacin del conjunto de tcnicas que constituyen el cdigo de barras en los
diferentes taxa de la vida se ha propuesto el uso de diversas regiones del ADN genmico,
principalmente secuencias ribosomales y del ADN mitocondrial (Raab y col., 2005;
Stockinger y col., 2010). No obstante, la aplicacin de estas herramientas moleculares sobre
el material biolgico provenientes de diferentes grupos de especies se enfrenta a diferentes
retos tecnolgicos para la correcta identificacin de grupos genticamente muy relacionados
(Chase y Fay, 2009). Por ejemplo, la determinacin a nivel especie en hongos (en el marco
del concepto de especie molecular), se basa principalmente en secuencias del ADN
ribosomal, como son la subunidad pequea (SSU), los espaciadores internos transcritos (ITS)
y la subunidad grande (LSU) (Stockinger y col., 2010). Sin embargo se ha observado que en

METODOLOGAS
DE
IDENTIFICACIN
EN
HONGOS

rdenes Eurotiales y Xylariales (phylum Ascomycota), las especies pueden ser identificadas
sobre la base de sus perfiles de metabolitos (Frisvad, 1994; Whalley y Edwards, 1995).
3.2.2. Ubiquinonas
Adems de los metabolitos secundarios, en taxonoma de hongos tambin se han utilizado
otros compuestos que desempean un rol esencial en el metabolismo, como es el caso de
las ubiquinonas (coenzima Q), stos compuestos desempean un papel importante en la
cadena del transporte de electrones del sistema respiratorio (Carlile y Watkinson, 1994). Se
sabe que el nmero de isoprenos adheridas al ncleo de la quinona es variable, y tales
diferencias se emplean para la clasificacin (gnero y subgnero) en levaduras; sin embargo
para las levaduras negras y hongos filamentosos el mtodo se ha utilizado de forma limitada
(Yaguchi y col., 1996) debido a que las conclusiones basadas solo en el sistema de
ubiquinona son debatibles, as mismo dependiendo del mtodo utilizado es posible obtener
diferente conjuntos de datos (Samson, 1991).

Conclusiones
El cdigo de barras del ADN como una propuesta novedosa para la identificacin a nivel de
especie de hongos, utiliza secuencias de ADN estandarizados para su aplicacin rpida,
precisa, universal y econmica, as mismo con el potencial de ser automatizado. Se han
propuesto varias regiones de ADN nuclear y mitocondrial para establecer un cdigo de
barras para hongos, sin embargo actualmente an no se ha definido una regin en particular
como se hicieron para los animales y plantas. Las regiones de ADN ribosomal ms utilizadas
para la identificacin a nivel de especie de hongos son los LSU, SSU, ITS, tambin se han
realizado estudios con COI con potencial para ser utilizada como cdigo de barras, sin
embargo se han observado limitaciones para algunos grupos de hogos en su aplicacin, por
lo que la creacin de un cdigo de barras universal para hongos se encuentra en desarrollo.
