Security
The SamplerSight-Pharma security system controls the following functions.
Who can log on.
What privileges they have (what they can do) while logged on.
Each users password, the minimum length of a password, and how long a password is
valid.
Whether SamplerSight-Pharma will automatically log off inactive users and how they
will be logged off.
Groups
Each Group represents a list of privileges. Each member of a Group will have all the
privileges of the Group. Groups can be added, modified and deleted if the current user has
that privilege. The Administrators Group cannot be deleted. A Group report is available
that shows all the configured Groups.
NOTE: Because individual users get their software privileges from the
Group they are a part of, the Groups should be created first.

Chapter 3 Security
Users
When SamplerSight-Pharma is first installed, a Default User and an Administrator user
are created in the Administrators Group. The Administrators Group has all privileges
enabled. Both users start without passwords, but they should be assigned a unique
password as soon as possible.
Each user belongs to a Group which determines the users privileges. In other words, a set
of privileges is set for each Group and any member of that Group has all the privileges of
the Group. Each user should be assigned a unique password. Users can be added,
modified and deleted if the current user has that privilege. Any user can be deleted with
the following exceptions:
The Default User cannot be deleted.
The last user in the Administrators Group cannot be deleted.
A user report is available that shows all the configured users.

In this chapter, Connecting refers to the steps necessary to enable the sampler to
communicate with the application and the sensor.
SamplerSight-Pharma supports the following RS-485 instruments:
Syringe
RS-485
NOTE: The word sampler refers to each and all of the listed samplers.
Prerequisites:
SamplerSight-Pharma has been correctly loaded on the PC.
One of the instruments listed above has been selected (see Chapter 2 Selecting the
Instrument).
Physical Connections
1 See the appropriate operators manual for the selected instrument to ensure all
connections have been correctly completed. For additional support, contact your
Particle Measuring Systems representative.

Sample Configuration RS-485 and RS-232
Once communication is established, the next major step is to define the sample
parameters. Each sample configuration is named and saved for future use.
When configuring a sample, entering information into the Sample Label fields is optional.
The program will prompt for information prior to initiating each sample, allowing
dynamic entry of variable identification data.
In cases where the information is static entering it during the initial sample configuration
stage will automatically populate the field each time the sample is run. An example of this
case would be if more than one instrument is owned, but they are located in different
areas. In this case it might be desirable to track the data generated by each instrument over
time to ensure consistent operator behavior. Entering the instrument serial number would
suffice, but using the actual physical location in the building might make it easier to
recognize which instrument generated the data.
Page 5-1
Page 5-2
When this check box is filled, the instrument/sensor samples continuously until the
Stop button is clicked.
Repeat Count
Repeat the sample x (integer) times. Ensure that Continuous Mode is not checked.
Discard Count
Sample Delay
The delay, in seconds, between samples. Set the field to zero (0) to have no delay
between samples. Sample delay is measured from the start of a sample to the start of
the next sample. This provides consistent start times. For example a set of data can be
created from samples starting every five minutes (300 seconds).
NOTE: If Sample Delay is less than the run time of the sample, no delay will occur.
If a dilution was required to reduce the counts below the coincidence limit of the
particle counter, enter the factor here. SamplerSight-Pharma will report all data as the
undiluted value of the sample. Entering one (1) will result in no dilution. Entering
zero (0) is not allowed.
Fill/Flow Rate (ml/min) The rate at which the sampler draws fluid through the sensor. This parameter is
specific to the sensor. Use the default unless there has been a clear directive no to do
so. The sensor is calibrated at a specific flow rate.
Syringe Volume (ml)
The volume of fluid remaining in the sample tubing. Can be calculated using formula
for volume of a cylinder or estimated at 1ml for the standard sample tube length
supplied with the instrument.
The actual volume drawn through the sensor. This volume can be greater than the
syringe volume. In these cases, the sampler will draw multiple strokes of the syringe
until reaching the sample volume.

Discard Count
If a dilution was required to reduce the counts below the coincidence limit
of the particle counter, enter the factor here. SamplerSight-Pharma will
report all data as the undiluted value of the sample. Entering one (1) will
result in no dilution. Entering zero (0) is not allowed.
This parameter is specific to the sensor. Use the default unless there has
been a clear directive not to do so. The sensor is calibrated at a specific
flow rate.

Limits
A user-defined limit can be set for the acceptable number of particles in a sample channel.
While limits are set and enabled, SamplerSight-Pharma will notify the user when the limit
has been reached in a sample.
All limits use the same normalization and summation selection that is entered in the
Limits tab in the Sample Configuration window (see Figure 6-3 on page 6-3). Each
limit has a specific channel, value type, comparison operator, and value. An unlimited
number of limits can be defined, and multiple limits can be defined for each channel.
Limits are defined in each sample configuration.

Fill the Display Limits check box in the Time Plot Properties window.
The limits will appear as a dotted line to differentiate them from the channel values.
Only the limits that match the currently selected summation and normalization will be
displayed. For example, if cumulative raw values are being displayed on the plot, then
only cumulative raw limits will be shown.

Display Configuration: Display, Labels,
Channels, Samples
The information in this chapter is relevant and common to any instrument supported by
SamplerSight.
Display configuration includes setting the following attributes:
whether the data displayed is Normalized or Raw
whether the data displayed is Differential or Cumulative
whether cumulative, normalized, or both are shown on the tabular display on the Main
Display window
the precision of the data
the appropriate data labels
the data channels that will be displayed and printed
how discarded data is handled
whether the data from each sample or each series will be automatically printed and what
report will be printed
what font and how many signature lines are used on selected reports
These attributes can be changed at the end of a series, but not during a series.
The data displayed on the monitor is the same as that which is printed.
To ensure that the correct data is reported, these settings must be correct before generating
a report.

Calibration
SamplerSight-Pharma includes a Calibration Wizard which guides the user through the
calibration process and enables the following operations:
Calibration of the sensor to all the particle sizes the sampler can detect
Verification of the calibration
View, print, or export the calibration data
Prerequisites
Training from a qualified Particle Measuring Systems representative
Near-monodispersed polystyrene latex (PSL) calibration particles of the sizes 2, 5, 10,
15, 20, 25, and 30 microns included in the Pharmaceutical Calibration Kit
Seven 100 mL containers with caps
200 mL container
Clean, solids-free detergent

follows:
a. Rinse the container thoroughly and leave the container filled with the last rinse
water.
b. Run the rinse water through the sensor to verify that no more than 100 particles ar
the smallest size of concern are present.
c. Repeat steps 3 4 until the sampler verifies that no more than 100 particles at the
smallest size of concern are present.
d. Cap the container.
3 Using Steps a., b., c., and d. in Step 2, clean and cap the remaining containers.
4 Discard the rinse water from a 100 mL container.
5 Add one drop from the appropriate PSL particle bottle to each sample container, fill the
container with 7590 mL of clean deionized or distilled water, and cap the container.
NOTE: Further dilution of smaller particles may be required, and greater
quantities (several drops) of larger particles may be required.
6 Repeat steps 89 until each bottle contains its appropriate PSL particles.

Nothing is downloaded to the sensor.
Finish
Available only if a previous calibration process was interrupted before all particle sizes
were processed. The Finish option allows resumption of the calibration process.
See Stopping Calibration before Completion on page 12-5.
3 Fill the Full radio button in the Calibration Wizard window, then click the Next button.
The Sample Configuration page of the Calibration Wizard window appears (see Figure
12-1).

The initial values shown on the size selection page will be the last values stored in the
calibration database for the selected flow rate. If no stored values exist for the selected
flow rate, then the values from the instruments calibration string are used, if one exists
and it matches that of the selected flow rate. Otherwise, predefined default values are
used.
Change the values on the Sample Configuration step only if you are certain they should
be changed.

The Calibration Wizard is minimized during the run to enable display of the histogram
and tabular data. When the calibration sample is complete, the Calibration Wizard is
displayed again.
NOTE: Once a size has been run the user can no longer click the
Back button to change the sample configuration.

11 When calibration data for the current particle size has been successfully collected,
move the sample intake tube to the next appropriate sample bottle, fill the check box for
that particle size, and click Next.
12 When all sizes have been run successfully, click the Finish button to end the calibration
process.
The calibration is stored to the calibration database, and the user is asked if they want to
download the calibration data to the instrument.
During download, a window announces that the data is being downloaded, and when the
download is complete.
13 Click the Finish button again to close the Calibration Wizard.

Verify Calibration
Verify Calibration
Verifying calibration allows the user to see how the calibration has changed since the last
calibration.
>> To verify calibration:
1 From the main display, click Perform > Calibration.
The Calibration Wizard appears.
2 Click the Verify option in the Calibration Wizard.
3 Click the Next button.
On the size page, a Size Change row will be displayed instead of raw Cumulative
Counts. Size Change is calculated as the percent size shift between the known actual
calibration particle size and the observed result size of the calibration sample. If the
calibration particle size is greater than the observed result size, the calculation is the
following:
(Actual Size - Result Size) / Result Size * 100
If the calibration particle size is less than the observed result size, the calculation is the
following:
(Result Size - Actual Size) / Result Size * 100

Calibration data can be selected and then sent to the instrument by clicking the Download
button. A window displays the status of the download. When the download completes, a
success message is displayed.
The download may take up to a minute to complete. If a communication problem occurs
before download completion, the download will never complete and no success message
will be displayed. In this case, the communication problem must be fixed and the
download attempted again.
The calibration report is generated by Crystal Reports. The report can be displayed,
printed, or exported.
