Abstract
Crocus sativus is a perennial sterile plant which is traditionally propagated via corms. Micropropagation of this
worthy plant by tissue culture through somatic embryogenesis is a rapid economic way to produce healthy and
pathogen free plant material. In the present study, somatic embryos of saffron were cultured on media containing
half strength MS salts with different concentrations (3%, 6% and 9%) of carbohydrates such as Sucrose, Glucose
and Maltose for 30, 60 and 90 days. All treatments were performed in complete random design each containing 4
replicas in 16/8h light/dark condition at 241c. Results showed that somatic embryos were best proliferated in
medium including Sucrose or glucose in 3% concentration for 90 days. Period of cultures and carbohydrate
concentration had a significant effect (p<0.05) on maturation of somatic embryos and cormlet production. The
highest numbers of embryos were converted to cormlet using medium with 3% concentration of carbohydrate
after 90 days. In comparison with other carbohydrates, sucrose was more effective for cormlet production.
