Intended use
The INNO-LIA HCV Score is a Line Immuno Assay (LIA), for the detection
of antibodies to human hepatitis C virus in human serum or plasma.
It is intended for use as a supplementary test on human serum or plasma
specimens found to be reactive using an anti-HCV screening procedure.

Test principle
The INNO-LIA HCV Score is a 3rd generation line immunoassay which
incorporates HCV antigens derived from the core region, the E2 hypervariable
region (HVR), the NS3 helicase region, the NS4A, NS4B, and NS5A regions.
The antigens were coated as 6 discrete lines on a nylon strip with plastic
backing. In addition, four control lines are coated on each strip: streptavidin
control line, 3+ positive control (anti-human Ig), 1+ positive control (human IgG),
and the cut-off line (human IgG).
The INNO-LIA HCV Score is based on the principle of an enzyme
immunoassay. A test sample is incubated in a trough together with the LIA test
strip. If present in the sample, HCV antibodies will bind to the HCV antigen
lines on the strip. Subsequently, an affinity-purified alkaline phosphataselabelled goat anti-human IgG (H+L) conjugate is added and reacts with specific
HCV antigen/antibody complexes, if previously formed. Incubation with the
enzyme substrate produces a chestnut-like color, the intensity of which is
proportionate to the amount of HCV-specific antibody captured from the sample
on any given line (Fig. 1). Color development is stopped with sulfuric acid.
Reagents
Description, preparation for use and recommended storage conditions

If kept at 2 - 8C, opened or unopened, all reagents are stable until the
expiration date. Do not freeze reagents.
All reagents and the plastic tube containing the test strips must be brought
to room temperature (18 - 25C) approximately 30 minutes before use and
returned to the refrigerator (2 - 8C) immediately after use.
Alterations in the physical appearance of kit reagents may indicate
instability or deterioration.
After opening the original tube containing the strips, any unused strip
will be stable for 16 weeks if stored at 2 - 8C in the closed original tube
with desiccant.

Test procedure
Please read Remarks and precautions before performing the test.

Use the required amount of test troughs, taking into account that for
each test run, a Positive and a Negative Control should be assayed.
Identify test troughs as controls and specimens, and place them in the tray.
Add 1 ml of Sample Diluent to each specimen test trough.
Add 10 l of the appropriate specimen or control to the appropriately
labelled trough.

Remove the required amount of LIA test strips from their container, and
add one strip to each of the test troughs. The test strip is placed membrane
side upwards into the trough using tweezers.
THE STRIP MUST BE COMPLETELY SUBMERGED.
Cover the troughs with an adhesive sealer (see Remarks and precautions).
Incubate the samples by placing the tray on a shaker or rocker (see
Direction for incubation) and agitate OVERNIGHT (16 2 h) at room
temperature (18 - 25C).
NOTE:
For the "2 hours and 3 hours sample incubation" protocol the same 15 steps
as for the test procedure "16 hours sample incubation" will be followed, but
changes to steps 3 - 5 - 6 and 9 have to be taken into account. Sample
volume for specimens and controls will increase from 10 to 20 l (step 3) and
sample incubation time changes to 2 and 3 hours (step 5). Washing after
sample incubation changes for the 2 hours procedure to 3 times 10 minutes
and for the 3 hours procedure to 3 times 6 minutes (step 6); finally the second
washing is 3 times 3 minutes for the 2 and 3 hours sample incubation.

Make sure not to damage the surface of the LIA test strips when
aspirating.
All the incubation steps (sample, conjugate, substrate, and sulfuric acid
incubation) and also the washing steps should be performed on a
shaker or rocker (use rocker only for overnight sample incubation).
During incubation and washing steps, the strip surface should be
completely submerged, with the membrane side facing upwards.
The shaker or rocker should allow a reciprocal (to- and- fro) motion of
the strips in the trough, and a movement of the liquid over the strips
without spilling over the trough.

For the "3 hours sample incubation" protocol the same 27 steps as for the
test procedure "2 hours sample incubation" will be followed, but changes to
steps 4 - 6 - 8 and 10 have to be taken into account. Sample incubation time
changes to 180 minutes for the 3 hours procedure (step 4) and washing after
3 hours sample incubation changes to 3 times 6 minutes (steps 6 - 8 - 10).

A reactivity rating must be made separately for each strip. To facilitate the
interpretation, a reading card is enclosed. The alignment of the developed
strip with the 3+ control line on the reading card makes the identification of
the lines easier.

Validation
Check the validity of the positive and the negative control strips and the
validity of the control levels on each strip before reading the test results.
Validation of the test run:
1. The positive control strip must show a reaction of at least 1+ on C1, C2,
NS3 and NS4 antigen line. The E2 and NS5 antigen line may show a
negative rating.
2. The negative control strip must show a negative rating (no reaction at
all or at least less than control level for all the HCV antigen lines).
Validation of a single strip:
1. The control levels 1+ and as well as the strong positive control level
3+ should be visible.
2. The intensity of the control level 3+ should be greater than that of level
1+ and the intensity of the level 1+ should be greater than that of level .
3. The control line for anti-streptavidin should have a negative rating.
NOTE:
Do not place paper on top of the strips as long as they are wet.

Seroconversion panels/low-titer panels
The results of the INNO-LIA HCV Score using the Auto-LIA 2-hour
sample incubation procedure and the manual 16-hour sample incubation
procedure were obtained internally on 13 BBI seroconversion panels (PHV
904 till 916), on 2 BBI low titer panels (PHV103 and 204), and on the
SFTS94 panel. These results were compared with CHIRON RIBA HCV
3.0 SIA and INNO-LIA HCV Ab III update results.
INNO-LIA HCV Score using the 2-hour and the 16-hour sample incubation
procedure detected antibodies earlier than CHIRON RIBA HCV 3.0 SIA in
7 panels, and 4 panels became positive at the same time. CHIRON RIBA
HCV 3.0 SIA detected antibodies earlier than INNO-LIA HCV Score using
both sample incubation procedures in one panel. One panel did not become
positive with any of these tests.
The INNO-LIA HCV Score using the 2-hour procedure detected antibodies
earlier than INNO-LIA HCV Ab III update in 2 panels, and the other 10 panels
became positive at the same time. Using the 16-hour procedure, 4 panels

HCV Ab IV. One of these 2 samples was tested on CHIRON RIBA HCV 3.0
SIA as well, and was found to be negative. For this sample set, an initial
specificity of 92.2% (189/205) was obtained, while specificity after repeated
testing was 93.7% (192/205).
Potentially interfering samples
One hundred thirty-seven potentially interfering samples were tested
internally using the manual 16-hour sample incubation procedure.
One hundred twenty-seven samples turned out to be negative, 9 were
indeterminate, and one scored positive. Upon repeated testing in duplicate,
this sample scored positive once, and then indeterminate, while a positive
result was obtained on INNO-LIA HCV Ab III Update. This sample was
found to be negative on Ortho HCV 3.0 ELISA with Enhanced SAVe, on
INNOTEST HCV Ab IV, and on CHIRON RIBA HCV 3.0 SIA.
On this set of samples, a specificity of 92.7% (127/137) was obtained.
Reproducibility
Two experimenters tested a panel of 13 HCV-positive samples, as well as
one positive and one negative control on 3 different lots using the Auto-LIA
2-hour sample incubation procedure while a third experimenter tested this
panel on one of these lots. The use of different strip lots and performance by
different experimenters resulted in the same test outcome.

Le test INNO-LIA HCV Score est un test immunoenzymatique sur bandelette
= Line Immuno Assay (LIA) destin la dtection des anticorps dirigs contre le

est destin tre utilis titre complmentaire, comme test plus spcifique, sur
des chantillons de srum ou de plasma humains qui se sont rvls positifs
dans le cadre d'une procdure de dpistage des anticorps anti-HCV.
Principe du test
INNO-LIA HCV Score est un test de 3me gnration de LIA qui utilise des
antignes HCV issus de la rgion Core, de la rgion hypervariable E2 (RHV),
de la rgion hlicase NS3 et des rgions NS4A, NS4B et NS5A.
Les antignes sont fixs sous forme de 6 fines bandes sur une bandelette
de nylon adhrant un support plastique. Par ailleurs, 4 bandes de contrle
sont fixes sur chaque bandelette: anti-streptavidine, Contrle Positif 3+
(anti-Ig humaines), Contrle Positif 1+ (IgG humaines), et la bande limite
(IgG humaines).

Le test INNO-LIA HCV Score est bas sur le principe d'un test
immunoenzymatique. Un chantillon est incub dans un compartiment avec
une bandelette LIA. Les anticorps anti-HCV ventuellement prsents dans
l'chantillon vont se lier aux bandes d'antignes HCV fixes sur la
bandelette. Ensuite, un conjugu anti-IgG humaines (H+L) d'origine caprine
coupl la phosphatase alcaline est ajout et se fixe au complexe
antigne/anticorps prcdemment form. L'incubation avec le substrat de
l'enzyme produit une couleur marron dont l'intensit est proportionnelle la
quantit d'anticorps anti-HCV spcifiques lis aux bandes d'antignes.
Le dveloppement de la coloration est arrt par addition d'acide sulfurique.

Les rsultats ont t obtenus en interne avec INNO-LIA HCV Score sur
Auto-LIA-programme pour incubation des chantillons sur 2 heures- et en
technique manuelle avec incubation des chantillons sur 16 heures, vis vis
de 13 panels BBI de sroconversion (PHV904 916), de 2 panels BBI de titres
bas (PHV103 et 204) et du panel SFTS94. Ces rsultats ont t compars aux
rsultats obtenus avec CHIRON RIBA HCV 3.0 SIA et avec INNO-LIA
HCV Ab III Update.
INNO-LIA HCV Score -procdure d'incubation 2 heures et 16 heuresa dtect les anticorps anti-HCV plus tt que RIBA 3.0 pour 7 panels. 4 panels
ont prsent une positivit en mme temps. CHIRON RIBA HCV 3.0 SIA a
dtect les anticorps anti-HCV plus tt que INNO-LIA HCV Score (pour les
2 procdures d'incubation) pour 1 panel. Un panel n'a donn aucune ractivit
par aucun des 2 tests.
INNO-LIA HCV Score -procdure d'incubation sur 2 heures- a dtect les
anticorps anti-HCV plus tt que INNO-LIA HCV Ab III Update pour 2 panels,
10 autres panels devenant positifs par les 2 tests en mme temps.
Avec la procdure d'incubation sur 16 heures, 4 panels sont devenus positifs
plus tt qu'avec INNO-LIA HCV Ab III Update, 7 panels sont devenus
positifs en mme temps et 1 panel est devenu positif plus tard.
Des 40 chantillons des 2 panels BBI de titres bas, 28 et 32 chantillons
respectivement ont t positifs avec INNO-LIA HCV Score -procdure
d'incubation 2 heures et 16 heures-. 35 de ces chantillons taient positifs
avec CHIRON RIBA HCV 3.0 SIA tandis que 34 l'taient avec INNO-LIA
HCV Ab III Update. Tous les chantillons positifs avec CHIRON RIBA HCV
3.0 SIA ou INNO-LIA HCV Ab III Update ont t trouvs positifs ou
indtermins avec INNO-LIA HCV Score.
Des 49 chantillons du panel SFTS94, 38, 41 et 40 respectivement ont t
positifs avec INNO-LIA HCV Score -procdure d'incubation 2 heures-, procdure d'incubation 16 heures- et INNO-LIA HCV Ab III Update. 31 de

Nehmen Sie fr jede Probe und jede Kontrolle je einen Teststreifen aus
dem Behlter. Legen Sie je einen Streifen in eine Inkubationswanne.
Die Teststreifen mssen mit der membranbeschichteten (markierten)
Seite nach oben in die Inkubationswanne gelegt werden. Sie drfen nur
mit einer Pinzette berhrt werden.
DIE STREIFEN MSSEN VOLLSTNDIG IN DIE FLSSIGKEIT

Mit der 2-Stunden-Methode wies INNO-LIA HCV Score bei 2 Panels Antikrper
frher als INNO-LIA HCV Ab III update nach, whrend der Nachweis bei 10
Panels zur gleichen Zeit positiv wurde. Mit der 16-Stunden-Methode gelang der
Nachweis mit INNO-LIA HCV Score bei 4 Panels frher. Bei 7 Panels wurden
die Proben zur selben Zeit positiv, und bei einem Panel spter.
Die zwei 2 BBI-Niedrigtiter-Panels enthalten 40 Proben, von denen jeweils 28
bzw. 32 Proben mit INNO-LIA HCV Score, 2-Stunden- bzw.16-StundenMethode, als positiv befundet wurden. 35 dieser Proben wurden mit
CHIRON RIBA HCV 3.0 SIA als positiv bestimmt, 34 wurden mit INNOLIA HCV Ab III update als positiv bestimmt. Alle Proben, die mit CHIRON
RIBA HCV 3.0 SIA oder INNO-LIA HCV HCV Ab III update als positiv
befundet wurden, waren mit INNO-LIA HCV Score positiv oder fraglich.
Von den 49 Proben des SFTS94-Panels waren 38 (INNO-LIA HCV Score,
2-Stunden-Inkubation), 41 (INNO-LIA HCV Score, 16-Stunden-Inkubation)
und 40 Proben (INNO-LIA HCV Ab III update) positiv. 31 dieser Proben
wurden mit CHIRON RIBA HCV 3.0 SIA als positiv befundet.
HCV-positive Proben
Insgesamt wurden intern 257 Proben von HCV-infizierten Patienten, die
zuvor mit 2 Screening-Assays und INNO-LIA HCV Ab III update als positiv
befundet wurden, mit INNO-LIA HCV Score (Auto-LIA 2-StundenInkubation) untersucht. Bis auf eine fragliche Probe wurde alle Proben als
positiv befundet. Daraus ergibt sich bei Einschluss der fraglichen Probe eine
Sensitivitt von 100% (257/257).
Unterschiedliche Genotypen
Intern wurden folgende Genotypen mit INNO-LIA HCV Score (Auto-LIA
2-Stunden-Inkubation) untersucht: 1a (15 Proben), 1b (15 Proben), 2 (15
Proben), 3a (15 Proben), 4a (15 Proben), 4 non-a (5 Proben), 5 (5 Proben).
Bei allen Proben fiel das Ergebnis positiv aus. Daraus ergibt sich eine
Sensitivitt von 100% (85/85).
Spezifitt
Blutspender
400 als negativ bewertete Blutspenderproben wurden intern analysiert (manuell,
16-Stunden-Inkubation). Beim Ersttest waren 377 Proben negativ, 22 fraglich
und eine positiv. Daraus ergibt sich eine Spezifitt von 94,3% (377/400).
Bei einem Wiederholungstest (Doppelbestimmung) wurde die zuerst als positiv
ermittelte Probe negativ befundet; die Spezifitt lag nun bei 94,5% (378/400).

Klinische Proben
205 klinische Proben wurden intern getestet (manuell, 16-Stunden-Inkubation),
davon 189 mit negativem, 11 mit fraglichem und 5 mit positivem Ergebnis.
3 der 5 positiven Proben wurden bei Neutestung als Doppelbestimmungen als
negativ befundet. Eine der zwei positiven Proben zeigte bei der
Wiederholungsbestimmung und auch bei INNO-LIA HCV Ab III update ein
positives Ergebnis. die andere Probe wurde mit beiden Tests als fraglich
befundet. Bei Ortho HCV 3.0 ELISA mit Enhanced SAVe und INNOTEST
HCV Ab IV war das Ergebnis negativ. Eine der beiden Proben wurde auch mit
CHIRON RIBA HCV 3.0 SIA getestet, ebenfalls mit negativem Ergebnis.
Die aus dieser Probenreihe ermittelte Spezifitt liegt bei Ersttestung bei
92,2% (189/205) und im Wiederholungstest bei 93,7% (192/205).
Proben mit potentiell zu erwartenden Wechselwirkungen
137 Proben mit potentiell zu erwartenden Wechselwirkungen wurden intern
getestet (manuell, 16-Stunden-Inkubation): 127 Proben waren negativ, 9 fraglich,
1 Probe war positiv. Bei Neutestung als Doppelbestimmung wurde einmal ein
positives und einmal ein fragliches Ergebnis erhalten. Mit INNO-LIA HCV Ab III
Update wurde mit dieser Probe ein positives Ergebnis erhalten. Bei Testung mit
Ortho HCV 3.0 ELISA mit Enhanced SAVe, INNOTEST HCV Ab IV und
CHIRON RIBA HCV 3.0 SIA war die Probe negativ.
Die aus dieser Probenreihe ermittelte Spezifitt liegt bei 92,7% (127/137).
Reproduzierbarkeit
Zwei Laboranten untersuchten ein Panel mit 13 HCV-positiven Proben
sowie drei verschiedene Chargen der Positiv- und Negativkontrolle mit AutoLIA, 2-Stunden Inkubation. Ein dritter Laborant testete das Panel mit einer
der Chargen. Trotz unterschiedlicher Chargen erhielten alle drei Laboranten
identische Ergebnisse.

INNO-LIA HCV Score un test immunoblot su striscia (Line Immuno Assay),

siero o plasma umani. Il suo uso previsto come test supplementare su
campioni di siero o plasma umani trovati reattivi utilizzando una procedura di
screening per anti-HCV.
Principio del test
INNO-LIA HCV Score un immunoblot su striscia di 3 generazione che
incorpora antigeni HCV derivati dalla regione del core, dalla regione
ipervariabile E2 (HVR), dalla regione elicasi, dalle regioni NS3, NS4A,

Gli antigeni sono stati immobilizzati in sei bande su strisce di nylon adese
ad un supporto plastico. In aggiunta sono fissate su ogni striscia quattro
bande di controllo: la banda di controllo della streptavidina, un controllo

INNO-LIA HCV Score basato sul principio di un test immunoenzimatico.
Un campione diluito incubato in una vaschetta insieme ad una striscia LIA.
Se presenti nel campione, gli anticorpi HCV si legheranno alle bande
antigeniche HCV presenti sulla striscia. Successivamente viene aggiunto un
coniugato, purificato per affinit, di fosfatasi alcalina legata ad anti IgG
umane di capra (H+L) che reagisce con i complessi specifici HCV antigene/
anticorpo, se precedentemente formati. L'incubazione con l'enzima substrato
produce un colore marrone, la cui intensit proporzionale alla quantit di
anticorpi HCV-specifici catturati dal campione su ogni banda specifica (Fig. 1).
Lo sviluppo del colore viene bloccato con acido solforico.

Muestras clnicas
Se probaron internamente 205 muestras clnicas utilizando el protocolo de
incubacin de muestra durante 16 horas. De ellas, 189 fueron negativas,
11 fueron indeterminadas y 5 resultaron positivas. Tras la repeticin del
ensayo por duplicado, 3 de estas 5 muestras resultaron negativas. Una de las
otras 2 muestras result positiva tras la repeticin del ensayo en INNO-LIA
HCV Score y tras el ensayo en INNO-LIA VHC Ab III Update, mientras que
la otra muestra result indeterminada en estos ensayos. Ambas muestras
fueron negativas en el Ortho VHC 3.0 ELISA con Enhanced SAVe y en el
INNOTEST VHC Ab IV. Una de estas 2 muestras fue probada tambin en
el CHIRON RIBA VHC 3.0 SIA y result negativa. Para este conjunto de
muestras se obtuvo una especificidad inicial del 92,2% (189/205), mientras
que la especificidad tras la repeticin del ensayo fue del 93,7% (192/205).
Muestras potencialmente interferentes
Se probaron internamente 137 muestras potencialmente interferentes
utilizando el protocolo de incubacin de muestra durante 16 horas. De ellas,
127 resultaron negativas, 9 fueron indeterminadas y 1 result positiva.
Tras la repeticin del ensayo por duplicado, esta muestra result positiva
una vez e indeterminada a continuacin, mientras que se obtuvo un
resultado positivo con el INNO-LIA VHC Ab III Update. Esta muestra
result negativa en el Ortho VHC 3.0 ELISA con Enhanced SAVe, en el
INNOTEST VHC Ab IV y en el CHIRON RIBA VHC 3.0 SIA. En este
conjunto de muestras se obtuvo una especificidad del 92,7% (127/137).
Reproducibilidad
Dos investigadores probaron un panel de 13 muestras VHC-positivas, as
como un control positivo y uno negativo, en 3 lotes diferentes utilizando el
protocolo de incubacin de muestra durante 2 horas con Auto-LIA.
Mientras tanto, un tercer investigador prob este panel en uno de estos
lotes. El uso de diferentes lotes de tiras y con ejecuciones por parte de
diferentes investigadores dieron lugar al mismo resultado de ensayo.

O INNO-LIA HCV Score um imunoensaio em linha (LIA) para detectar
anticorpos para o vrus da hepatite C humana em soro ou plasma humano.
Deve ser utilizado como teste suplementar em amostras de soro ou plasma
humano que tenham sido reactivas com um procedimento de despiste anti-HIV.

Princpio do teste
O INNO-LIA HCV Score um imunoensaio em linha de terceira gerao,
que incorpora antignios HCV derivados da regio nuclear, a regio E2
hipervarivel (HVR), a regio NS3 helicase, as regies NS4A, NS4B, e NS5A.
Os antignios esto impregnados em 6 bandas distintas numa tira de nylon
sobre um suporte de plstico. Alm disso, foram impregnadas quatro bandas de
controlo em cada tira: banda de controlo de streptavidina, banda de controlo
positivo 3+ (Ig anti-humana), banda de controlo positivo 1+ (IgG humana) e
banda cut-off (IgG humana).
O INNO-LIA HCV Score baseia-se no princpio de um imunoensaio
enzimtico. A amostra de teste incubada numa cavidade juntamente com a
tira de teste LIA. Se existirem na amostra, os anticorpos HCV ligam-se s
bandas de antignio de HCV da tira. Subsequentemente, adicionado um
conjugado de purificado de IgG anti-humana de cabra marcada com fosfatase
alcalina (H+L), que reage com complexos antignio/anticorpo especficos de
HCV, se formados previamente. A incubao com o substrato enzimtico
produz uma cor acastanhada, cuja intensidade proporcional quantidade
de anticorpo especfico de HCV capturada da amostra em cada uma das
bandas (Fig. 1). O desenvolvimento da cor parado com cido sulfrico.

Validao do teste:
1. A fita de controlo positivo deve apresentar uma reaco de pelo menos
1+ na linha de antignio C1, C2, NS3 e NS4. A linha de antignios E2 e
NS5 pode apresentar um ndice negativo.
2. A tira de controlo negativo deve mostrar um ndice negativo (nenhuma
reaco ou, pelo menos, reaco inferior ao nvel de controlo para
todas as linhas de antignio HCV).
Validao de uma tira nica:
1. Os nveis de controlo 1+ e e o controlo fortemente positivo 3+ devem
ser visveis.
2. A intensidade do nvel de controlo 3+ deve ser maior que a do nvel 1+
e a intensidade do nvel 1+ deve ser maior do que a do nvel .
3. A linha de controlo para anti-streptavidina deve ter um ndice negativo.

Painis de seroconverso/painis de baixo ttulo
Os resultados do INNO-LIA HCV Score com o procedimento de incubao
de amostra de 2 horas Auto-LIA e o procedimento manual de incubao de
amostra de 16 horas foram obtidos internamente em 13 painis
seroconverso BBI (PHV 904 - 916), 2 painis de baixo ttulo BBI (PHV103 e
204) e no painel SFTS94. Estes resultados foram com os resultados
actualizados do CHIRON RIBA HCV 3.0 SIA e do INNO-LIA HCV Ab III.
O INNO-LIA HCV Score com o procedimento de incubao de amostra de
2 horas e 16 horas detectou anticorpos mais cedo do que o CHIRON RIBA
HCV 3.0 SIA em 7 painis e 4 painis ficaram positivos em simultneo.
O CHIRON RIBA HCV 3.0 SIA detectou anticorpos mais cedo do que o
INNO-LIA HCV Score com ambos os procedimentos de incubao de
amostra num painel. Um painel no ficou positivo em nenhum destes testes.

Especificidade
Dadores de sangue
Foram analisadas 400 amostras de dadores de sangue negativas para
anticorpos HCV internamente com procedimento manual de incubao de
amostras de 16 horas. Depois do teste inicial, 377 amostras foram negativas,
22 amostras indeterminadas e 1 amostra positiva, resultando numa
especificidade inicial de 94,3% (377/400). A amostra de sangue positiva
inicial teve um resultado negativo depois da repetio do teste em duplicado,
resultando numa especificidade depois do teste de 94,5% (378/400).
Amostras clnicas
Foram testadas duzentas e cinco amostras clnicas internamente com o
procedimento manual de incubao de amostra de 16 horas. Cento e
oitenta e nove amostras foram negativas, 11 indeterminadas e 5 positivas.
Trs destas 5 amostras tiveram resultados negativos depois da repetio do
teste em duplicado. Uma das outras 2 amostras positivas tiveram
resultados positivos depois da repetio do teste no INNO-LIA HCV
Score e no INNO-LIA HCV Ab III Update, enquanto a outra amostra teve
um resultado indeterminado nestes ensaios. Ambas as amostras foram
negativas no Ortho HCV 3.0 ELISA com Enhanced SAVe e no
INNOTEST HCV Ab IV. Uma destas 2 amostras foi testada no CHIRON
RIBA HCV 3.0 SIA e teve um resultado negativo. Para este conjunto de
amostras, foi obtida uma especificidade de 92,2% (189/205), enquanto a
especificidade depois da repetio do teste foi 93,7% (192/205).
Amostras potencialmente interferentes
Foram testadas cento e trinta e sete amostras potencialmente interferentes
internamente com o procedimento manual de incubao de amostra de
16 horas. Cento e vinte e sete amostras foram negativas, 9 indeterminadas
e 1 positiva. Depois de repetir o teste em duplicado, esta amostra teve um
resultado positivo e, em seguida, indeterminado, enquanto um resultado
positivo foi obtido no INNO-LIA HCV Ab III Update. Esta amostra teve um
resultado negativo no Ortho HCV 3.0 ELISA com Enhanced SAVe,
INNOTEST HCV Ab IV e no CHIRON RIBA HCV 3.0 SIA. Neste
conjunto de amostras, foi obtida uma especificidade de 92,7% (127/137).
Reprodutibilidade
Dois tcnicos testaram um painel de 13 amostras de HCV positivo, um
controlo positivo e um controlo negativo em 3 lotes diferentes com o
procedimento de incubao de amostra de 2 horas Auto-LIA enquanto um
terceiro tcnico experimentou este painel num destes lotes. A utilizao de
diferentes lotes de tiras e o desempenho de diferentes tcnicos resultaram
no mesmo resultado de teste.
