Abstract
The classical me thod for counting plate le ts (Thrombocytes)
is use of two types of microscopy: Burcker Turk chambers
(hemacytome te r) and slides with pe riphe ral blood smears.
In the last te n years, a ne w me thod, flow cytome try with
application of the impe dance principle has bee n de ve lope d
for CBC and counting of plate le ts.
Scope :
The current studies sought to compare the results of plate le t
counts from the optic microscope with the values obtaine d
by use of flow cytometry and the impe dance principle in
samples obtaine d from patie nts be fore and after unde rgoing
hemodialysis.
Me thods:
Three methods were use d to assess plate le t counts of
hemodialysis patie nts: optical microscopy, pe riphe ral blood
smear and use of the cytome try principle with impe dance
principale (VIC) by Coulter HNX he matological analysis.
The study was performe d on 90 patie nts, 55 men and 35
wome n, ages 35-65 years (mean age 50), hospitalize d ove r a
3 month pe riod. The patie nts we re analyze d once monthly,
all at the same day, before and after dialysis.
The sample s we re assesse d for plate le t count by statistical
paramete rs: [SD =(Xi- Xm)2 /n 1; Acuracy: (%Diff = X
average X
targe t/ X mean x 100, with normal value until + 25) and Z
score( Z = X ave rage-Xtarget/SD, with normal value until +-2,
R>0.95%), for ave rage plate le ts 150-400 x10/l, 95% CI.].
Results:
The comparison be tween the plate le t counts on the Coulter
HMX (mean value X = 233 x 10l; p=0.028; SD=2; %
Diff=0.90; Z score = - 0.30) and by optical microscopy (X =
250 x 10l; p=0.029; SD= 2.6; %Diff = -3.6; Z score =0.40)
yie lde d similar values in a control group (120 male and
female healthy subjects, ages 25-55 years( mean age 40). A
similarity betwee n results for the me thods was also found
for the plate let counts of hemodialysis patie nts assesse d
Introduction
In order to avoid
systematic errors during
platelets(Thrombocytes) count by optical microscopy,
a method of direct counting in the Burker-Turk
chamber( hemacytometer) has been recommended for
use in parallel with determination of the number of
Thrombocytes counted on peripheral blood smear,
( by optical microscopy). Calculation of the platelets
counted in the Burker-Turk chamber considers the
height of the chamber and the surface of the middle
