PLASENTA MENCIT (Mus Musculus) MODEL PREEKLAMPSIA

The Influence of L Arginine on endothelial damage in Preeclampsia Model of Mices


Placenta (Mus Musculus)

ABSTRAK
Introduction : Preeclampsia is one of the primary contributor for morbidity and mortality in pregnant
women and fetus. In preeclampsia, increased advanced glycation end products (AGEs) and their receptors
can be identified. This process will activate the Nicotinamide Adenine Dinucleotide Phosphate (NADPH)
oxidase to support the expression of souble Fluid-like tyrosine-1 (sFlt-1) in trophoblasts which are
associated with increased oxidative stress, especially in the placenta. In this regard, L-arginine plays a
significant role in the maintenance of preeclampsia, through the nitric oxide synthase (NOS) pathway. L-
Arginine inhibits NOS inhibitors, hence it increases the production of NO, promotes vascular
vasodilatation and improves hypoxia and reduces oxidative stress. This reserach aims to learn the effects
of L-ARGININE administration to endothelial damage in preeclampsia model of mices placenta ( Mus
Musculus).
Methods : This research was an experimental study with 30 pregnant mice (Mus Musculus), divided
randomly into three groups, normal pregnancy, preeclampsia model and preeclampsia model given L-
Arginine. Termination of birth was done in 16th day, the changes in histological features of placenta on
the villi chorialis artery thickness and diameter were observed. Statistical analysis of the histology arterial
diameter was done using Kruskall-Wallis statistical test. After the Kruskall-Wallis test the data would be
analyzed using the Mann Whitney test. The statistical analysis of the histologic thickness of villi chorialis
artery was done using one-way ANOVA statistical test. After the one-way ANOVA statistical test the data
would be analyzed using the post-hoc t test.
Results : The mean of villi chorialis arterial diameter of normal pregnant mice was 370,54 m compared
to the mean of villi chorialis arterial diameter of preeclampsia model was 309,03 m with p=0,029
(p<0,05). The mean of villi chorialis arterial diameter of preeclampsia model was 309,03 m compared to
the mean of villi chorialis arterial diameter of preeclampsia model given L-Arginine was 365,25 m with
p=0,015 (p<0,05). The mean of villi chorialis arterial diameter of normal pregnant mice was 370,54 m
compared to the mean of villi chorialis arterial diameter of preeclampsia model given L-Arginine was
365,25 m with p=0,648 (p>0,05). The mean of villi chorialis arterial thickness of normal mice was 50,95
m compared to the mean of villi chorialis arterial thickness of preeclampsia model was 70,52 m with
p=0,009 (p<0,05). The mean of villi chorialis arterial thickness of preeclampsia model was 70,52 m
compared to the mean of villi chorialis arterial thickness of of preeclampsia model given L-Arginine was
51,56 m with p=0,011 (p<0,05). The mean of villi chorialis arterial thickness of normal mice was 50,95
m compared to the mean of villi chorialis arterial thickness of preeclampsia model given L-Arginine
51,56 m with p=0,931 (p>0,05).
Kesimpulan: Pemberian L-Arginine terbukti memberikan perbaikan kerusakan endotel pada plasenta
mencit (Mus Musculus) model preeclampsia.
Conclusion : The administration of L-Arginine was proven to improve endothelial damage in
preeclampsia model of mices placenta (Mus Musculus).
