Millons Add Millons reagent [Hg(SO4) + HNO3] Red to pink color phenolic amino acid tyrosine

test tube. Mix thoroughly. Then add 1 ml
of concentrated sulphuric acid, pouring it
down along the side of the test tube

Reduced d i f f e r e n t tubes with 1mL each of 10% NaOH Brown to black sulfur containing amino acids
Sulfur and saturated lead acetate, PbAc precipitate cysteine and cystine
Boil in a water bath for
3 minutes

Sullivan 1 mL of protein solution containing Red color Cysteine or cystine


Cystine Cysteine/Cystine in a test tube, add 3
drops of a 5% solution of sodium
nitroprusside

Sakaguchi To 1 mL of protein solution in a test tube, Red color Guanidine group in arginine
add 40%NaOH (2-4 drop) + Ethanolic a-
Naphthol (2 drops) and Bromine water (5-
10 drops)

Diacetyl Mixing a dilute solution of the protein or Pink color Guanidine group
other substance in question with a small
quantity of 100/0 KOH and then adding
one drop of a 10/o solution of diacetyl.
Nitroprusside To 1 mL of protein solution containing Red to purple color Proteins containing sulfur , -SH
Cysteine/Cystine in a test tube, add 3 (in cysteine & cystine)
drops of a 5% solution of sodium
nitroprusside - Mix well and add few
drops of ammonia solution

Bromine add 3 drops of bromine water and 1mL of Pink color Pyrrole ring of tryptophan
Water n-amyl alcohol.Shake well. o b s e r v e
the colour of the alcohol
