Materials and Methods

I. Materials:
Beaker pH paper
Pipette Acetic acid (HOAc)
Aspirator Sodium acetate (NaOAc)
Test tubes Sodium hydroxide (NaOH)
Test tube rack Hydrochloric acid (HCl)
Dropper Phenolphthalein
Centrifuge Methyl orange
Syringe Blood serum
Redtop vacutainer tube Distilled water

II. Methods
A. Preparation of Buffer Solution

Prepare a 0.1 M buffer solution using a 0.10 M


HOAc (acid) and a 0.10 M NaOAc (salt) solutions
with a pH of 4.4 and a total volume of 100ml.

Calculate for the volumes of the acid or base and


salt to be used.

Measure the calculated volumes using burettes.

Mix and check the pH using pH meter

If measured pH of your buffer deviates from the


assigned value by more than 0.1 add drop wise
either 0.1M HCl or 0.1 M NaOH to bring the
buffer to within +/- 0.05 of the desired pH value.

Record the original pH and the number of drops (if


any) of NaOH and HCl added.
B. Buffer action of Blood serum

Prepare the four solution in Extract blood and centrifuge it


four different test tube. in order to get the serum.

In test tube number 3, add 0.1 M In test tube number 4, add 0.1 M
NaOH dropwise until it matches HCl dropwise until it matches
the color of test tube number 1 the color of test tube number 2

Record the total number of drops of 0.1 NaOH


and 0.1 M HCl in each case
