How to collect dried blood spots on filter paper for malaria

research?
I need to collect dry blood spots on filter papers for future PCR analysis. I'll be collecting
the blood samples from patients visiting clinics in a community. What is the procedure?
How can I be sure of my sample size? What is the best method of storage?

Procedure for making dried blood spot is simpler, first we need to cut Whatman filter
paper (Number 3) into small circular or rectangular strips and autoclave it. Cover the filter
strips with a transparent film so that you we can avoid any cross contamination between
samples. You can put finger prick's blood at one, two or three position on filter strips. Dry
it completely at room temperature and store at 4 C for future use. We are using more than
6 year old dried blood spots and getting good results

I appreciate your response to my question, however I need more clarifications in some
areas. 1. The filter papers should be cut into what dimensions (please give an example).
2. Do I autoclave before placing drops of blood samples on the filter papers? 3. After
autoclaving, do I need to dry before using the filter papers? 4. Why do I need to autoclave,
is it to disinfects? I guess storage at 4 Celsius is when I had air-dried the blood sample
and place in plastic bags. If yes, do I need to place desiccants into the plastic bags
before storing at 4 C?

Hi Soniran I also recommend Whatman™ FTA™ for blood DNA collection. Attachment
please find the related information what you want.

FTA card is the best one but u can used what man 3 filter paper with fine dry and good
storage its more cheap and affordable

Yes, you correct but autoclaving also degrades DNAse. We never used desiccant for
storage but if you would like you can.

When you have the whatman filter paper cut sizeable part about 3cm size put a drop of
blood on the filter paper and let it dry and store in silica jell container untill ready to use
for pcr.

apply 2-3 drops of blood on a standard filter paper in duplicates (optional).allow it to air
dried, seal in a minigrip plastic bag and store at -20 for further molecular ab. use.
A sample of 72 is usually considered to be sufficient
