Each instrument has specific functions and methods. The user, while
consulting the manual, must consider only the Elfolab functions belonging to
the instrument in use.

6 Software Installation Elfolab

Software installation

Elfolab is supplied with CD-ROMs 700 Mb each of them labelled with the
title, the version and the revision number of the two software.

System Requirements
For a correct installation of Elfolab the following items are required:
 Personal Computer: Intel Core, Intel i3, i5 or more.
 RAM: 1 G Minimum.
 Screen: Minimum resolution of 1024x768 pixels; 64k Colors.
 Hard-Disk: 5Giga Minimum.
 Operating System Windows: Windows XP, Windows Vista,
Windows 7, Windows 8.

Power Options

In order to avoid problems on the connection between PC and instrument the user
should always set the PC to maximize its performance:
• The screen should never be turned off;
• Hard drives should never be disabled;
• The PC must never be idle;
• The PC must never be suspended.
It is possible customize the settings in the Control Panel-> Power Options.

Preliminary steps for Windows Vista


Users, who utilize Windows Vista, before installing the Elfolab software,
must remove some protections that block the regular usage of the programs.

Windows Defender Removal

1. Open Control Panel form Menu Start of PC at lower left.

2. Select the icon “Windows Defender”.

Preliminary steps for Windows 7/8

Users running Windows 7/8 before installing the software


Elfolab / Interlab G26, remove some protections that prevent the proper
program functions.
Removing User Account Control

1. Enter the Control Panel from the Start Menu low in the left.

3. Select Modify the settings of User Account Control.

Installation procedure

Note! In case of a re-installation or installation of an up-date version, it is necessary

to uninstall the previous version before loading Elfolab (cfr. Uninstall


Procedure pg. 19).

Elfolab Installation procedure


 Insert the CD-Rom “INTERLAB CD” in the CD drive.
 Select the icon “Open The folder to visualize the files...”.

 Select the folder “Elfolab x.x.x” inside the CD.

 Select the icon “Setup.exe”.

 Close all the applications and select “OK”.

 Choose the destination folder (it is recommended to select the predefined one)
and press the button with the PC icon.

Uninstall Procedure
1. Turn the computer on starts and the desktop will appear.
2. Select Start in the Application Bar
3. Select Control Panel.
4. In Control Panel double click on Add/Remove programs.
5. In the dialog box Add/Remove programs select Elfolab in the list of
applications, click on Add/Remove.
6. Answer Yes when you are requested to confirm the removal of Elfolab from
the computer.
7. A window asking whether or not maintaining shared files may appear at this
step. In this case, choose the Remove all option.
8. Windows will automatically remove the application files from the Hard Disk
as well as the references of the application of the files in the Windows
registers. The message showing the complete uninstall procedure will appear.
9. Click on OK to close the procedure.

The uninstallation of Elfolab doesn’t erase the Elfolab database.

Note!
Starting Elfolab
If the installation has been performed correctly, the icon of
Elfolab will be displayed in the Windows programs menu. To run Elfolab do as
follows:

1) Select Start in the Application Bar.


2) Select Programs.
3) Click on Elfolab icon.
When you run Elfolab for the first time after installation, the following
window will appear:

 The program will ask to choose the type of the instrument. Select
the instrument by clicking on the appropriate box.
 Depending on the instrument it is possible to choose a scanner for
the image acquisition of the “immunofixation” method.
 Moreover, as some instrument having the EEPROM version greater
or equal to 4.18 can choose the number of depositions, it is necessary
to select the EEPROM of the Tool in use.
 In order to use the automatic sampler ASD52 (only for Microgel or
INTERLAB G26) select the corresponding checkbox and then press
OK.

 When the Instrument has been selected push OK.

 The following window will appear:

 Press button to run Elfolab for the first time.

In case information regarding the number of records of patients is reported,


please note that the maximum number of patients that can be analyzed in one
full working cycle is listed below:

1 A new day of work

1.1 Running Elfolab

If the installation has been performed correctly, the icon of Elfolab will be
displayed in the Windows XP /Vista program menu. To run Elfolab do as follows:

1) Select Start in the Application Bar.


2) Select Programs.
3) Click on Elfolab icon.
4) If the selected instrument is Microgel or INTERLAB G26, the first
time that the program is started it will be asked for the directory where

the file Microgel.exe (icon ) or INTERLAB G26.exe (icon for

series 33 and icon Errore. Non si possono creare oggetti dalla

5) If the selected instrument is Pretty - ScanLab the first time that the
program is started it will be asked for the directory where the files
PrettSoft.exe and ScanLab.exe are installed.

It needs to click “OK”, to select the right folder and file where
PrettySoft.exe is installed and to click “Open”.

is installed and to click “Open”.

6) The software will show this window (the image is depending from
instrument selected):

1.2 Open the Database

To open the database you have to select Open Database in the Database
menu.

Database: A database is a list of information concerning patients. The

Note! database is divided in 2 areas: the historical and the working area. The data
are received in the working area; later it is possible to save after in the
historical area.

1.3 Inserting Patients Data

The database is empty. Insert new patient data as follows:

1) Click on or select Working Database in the Database menu;

2) In the following window:

select “New Working Database”.


3) Select the method for the corresponding session and click ok.

Otherwise the choice can be delayed to a following step. In this case simply select

the button for Microgel or for INTERLAB G26 and then the
remaining gel holders or session could be selected.
In such way empty records will be created and will be ready for the filling of the
remaining information.

The manual selections of methods will not more present for


the instrument Interlab G26 (series 36, 42 and 48) if the
Note! customer selects Sessions Management “Yes” in the menu
Settings->General Parameters (see page 88).

In case the last session data should be retrieved select

Warning!

A wrong choice can change the correlation between the patient graphs and the
relative data. If the next running method is not known in advance, in the Microgel, it is

3) In order to insert patient data, to refill the Patient Area Data. The user
can move from one field to the next using the TAB key on the
keyboard.

4) Then press the Enter key to go to the next patient record.

The first graph of the first gel holder will be assigned to the first patient, the
second graph of the first gel holder to the second patient and so on until the list of
patients is completed.

Record: Each record contains all the information and the eventual notes
Note! concerning the patient.

1.4 Importing and exporting data from file

If the system is connected with a central information system (i.e. laboratory


information system, LIS), it is possible to import and export data in an ASCII
format with the .asc extension.
The communication follows the Communication Protocol that is provided
from Interlab S.r.l.

 Data Import

1. Select File > Import Data or clicking on the icon:

2. Select Yes when asked if you wish to import data.

3. The following screen will appear. Select what files to upload and whether to
overwrite any existing information already inserted in the work list. Select
OK.

4. It is also possible to search the Host system for all patient demographics and
data or select specific samples for the search.

Once the program reads a list from the Host, the following summary
window is displayed:

The first column contains the Elfolab values before the import process.
The second column contains the record list coming from the Host that has
been coupled to the records already present in Elfolab.
The third column shows the Host list.
All the records that are going to create the future list, in case the import is
confirmed, are colored GREEN.
All the Host list records that have found a coupling with the Elfolab list are
colored YELLOW.
The coupling will not be done until the button “Import data” is pushed so
the list present in Elfolab will remain unchanged.
The buttons at the bottom of the window allow the following operations:
import data, exit without importing, print the Host list and the Host list of the not
matched records.
The matching of the data present in Elfolab and in the Host
list is realized only with the “Sample ID” field!

Warning!

These functions are available only if you are in the working database, which
is therefore ready for patient data entry.

 Data Export

Once the analysis is completed and results have been associated to patients,
it is then possible to send data containing the Sample ID, the result of the analysis
and the graph in the hexadecimal numeric format to the Central Information
System by the command Export data to file.

It is possible to use the command Export data to file from the menu File or

the icon .
In this case in the following mask the customer will can decide which data
to export.

It is possible to export the data still from the historical database. In this
case the export data will available only from menu File. It will possible to export
the current patient (Export Data Current Patient) or more records previous
filtered from the Filter (Export Filtered Data) .

 Immunofixations Export
It is now possible export the images of Immunofixations in Jpg format.
The commands are the same as the export data. The images will be exported in the
export folder.
In order to export the exporting Immunofixations:
1. In the menu Settings->General parameters the parameter
“Export Immunofixations Images” have to have “Yes”;

2. The Field Sample ID musts not be empty.

The name of image exported will be the field Sample ID. In the example the
name of file will be “371640.jpg”.
In case of Immunofixations method still the antiserum names will export.

1.5 Viewing the List of Patients

During the data entry, or after the data entry is completed, it is possible to

visualize a list of the patients by clicking on or by selecting Show List in


the File menu.

 In these windows you can edit or add data, save and print the list.
 In the Session Frame the user can select the sessions to display and
print.
 In the "Enter keyboard" Frame the user can decide how to advance
in the fields of the table by pressing the "Enter / Return".

What information is viewed and/or printed is determined by the user. To


select the desired fields, select Settings > Patient List Settings. The following
screen appears.

From here, the user can select fields to view and/or print on the work list.
Note: a maximum of 6 fields may be selected for printing.

Currently entered patient data are stored in the temporary database, and can
be stored permanently in the historical database by using the option Database >
Save last reading or by clicking on the icon, shown on the Elfolab main
window.

1.6 Interact with the Instruments

In order to connect the Instruments to the PC and to start the analysis, refer
to chapters 2 and 3 according to the Instrument type.

1.7 Value Area

Once the data have been received, Elfolab will show the graph, name and
value of the fractions both in % and concentration.

Furthermore a dedicated icon will immediately highlight any anomalies by


turning red. In this case the user could also observe in red color the difference
between the minimum and maximum of the normal value range and the
current value.

Finally, clicking on “Conc.” button, the user can see if the anomaly is
present in concentration too.

It is also possible to eventually change the name of a fraction directly from this
window by clicking on the relative text box.
In Chapter 4 it is possible to see how to edit names and default ranges of the
fractions for all the methods.

It is possible to analyze one or more samples of Urinary Proteins or CSF during


the Serumproteins analysis. Elfolab software allows, after data transfer, assigning
the right method to the processed sample by clicking on one of the two buttons at
the lower right of the main Elfolab window

1.8 Results printing

Push the icon or the item Print from the File menu to show all the
print possibilities. The following window will appear for the choice:

It is possible to print the results of the current selected patient or of a filtered


list of patients. It is also possible to print all the temporary patients’ results or just
an interval based on sequential number.
All print possibilities are shown in detail in paragraph 4.5.

2 Interact with Microtech and Genio

2.1 Connection with the instrument

The communication to the Instrument is set up through two


parameters: communication port (Com Port) and data transfer speed
between Instrument and Computer (Baud Rate).

1) Simply select the port or the desired baud rate. Note that once the
communication is active, the port cannot be changed.

2) In order to start the communication to the, click once on or select the


item Connect with Instrument from menu File.

If the communication starts correctly, in the message bar at the bottom of


the panel a confirmation message appears:

and also the symbol on the right of the message.


In the same bar error messages can be shown in case of problems. If this is
the case check the connection and consult the Instrument manuals.

Make sure the instrument is switched on before connecting to it!

2.2 Method selection

After the connection between the computer and the instrument has been
established, you can choose the type of the analysis you wish to run in the
Methods menu. Two types of methods are available, one for Methods with
reading and one for Methods without reading.

The methods listed in the picture above may vary according to the
type of instrument.
2.3 Control Panel

After the analysis has been chosen, a Control Panel window will appear.

Strips to be
analyzed

At the top part of the panel, the strip(s) to be analyzed can be selected by
simply moving the mouse pointer to the numbered box(es) and clicking
on it. A check mark will show up in the selected box.
Once the analysis is started, in the same way proceed to select in the
lower frame the analyzed strip(s) to be acquired.
Following the meaning of the commands:

Starts the analysis procedure

Pause the cycle (after completing the step);

Restart from the pause status;

It is used to read a strip already analyzed (i.e. the day before or


from archive) and it is necessary to specify the location of the
strips to be read;

It starts an internal control of the Instrument in order to detect any


problem.
At the end of the check a window can be shown with messages
about the problems and problem causes. If any problem is
detected it is necessary to solve them and then click

, then again on to start the analysis


of the strips automatically detected by the Instrument;

It starts, without any pause, a full Instrument cycle. It can be used


to verify the correct function of the Instrument that will
perform all the movements required for the analysis.

At the bottom area of the control panel, there are other buttons that allow
data transfer:

Starts, after selecting the strips to be received, the transfer


of data already analyzed for a new calculation;
Acquires again the already read data in case the calculus
parameters have been modified (operation done in Setup-
Calculus). It is enabled only if the data for the strips
involved in the calculus have been already received.

In addition, in the middle area of the window, it is shown if the


method is micro or semi-micro (cfr. paragraph 4.6).

N.B. For Genio S instrument, after selecting the


following window will appear. It’s possible to choose if the analyzed strip
is 4 or 8 traces:

2.4 Instrument Status

On the Control Panel there is the button. It allows


opening a window that provides a general overview of the status of the current
analysis and the settings. This window can be opened at any time during the
analysis.

The window shown above may display slight variations according to the
selected type of analysis.
This window can be opened during the simulation cycle also. It offers a
detailed view of the analytical cycle, from the strip soaking to the densitometric
scanning of the patterns on the strip.
All the steps of the selected method are listed on the left part of the window.
The position of the strip during the analytical process is displayed by the Interlab
logo on the reference column in the Show Status window
Current type and status of the analysis
and some parameters of the instrument are
shown on the right side.
In the Analysis area, the selected method
and the current status of the analytical process
are indicated by the green boxes. The Suspended box will turn green when the
instrument is completing the step in progress, before the analytical process is
suspended.

The Phase Timings to end area includes one


box showing the total time elapsed from the
beginning of the analytical cycle, and six numbered
rectangles, each referred to the associated strip
number. For each strip, the time lasting to the conclusion of the current step is
displayed.

This box indicates current values of voltage,


current, cooling temperature (of the Peltier plates)
and dryer temperature.

You can close the Show Status window by clicking on

2.5 Methods without reading

Strips to
analyze

1) The strips to be analyzed can be selected by simply moving the mouse


pointer to the box, in the upper part of the window, and clicking on it.

2) Clicking on will then start the analytical procedure.


3) If the computer is connected to a scanner (cfr. Starting Elfolab, pg.19),
it is possible to do the image acquisition, only for the Immunofixation
method, as explained below:
 Put the strips, obtained by the Immunofixation method, on the
glass plate of the scanner;

 Click on ;

 Select the type of scanner available on the following window:

 Run the scanning by clicking on ;

 Clicking this button will enlarge the scanned picture;


 If you wish to save the picture in the directory you choose by

using this button ;


 Once the acquisition is completed, you can return to the main

menu window by clicking on .

This button does not appear if no scanner is connected to the

computer (cfr. Starting Elfolab, pg.19).

4) In the main menu window it is possible to give each patient the


correspondent result of the analysis. Print menu allows printing the
reports.
In case a scanner has been selected during the software installation (cfr.
First Run, see page 19), the image of a strip can appear in the Graph Area of the
Results (see. Page 23) after the analysis:
 If a scanner is not available, in the Graphic Area of Results an image
will appear that represents a strip. After visual inspection of the strip,
click on the boxes corresponding to the identified bands.
 If a scanner was used for data acquisition, 6 new patient records
were created; each of them can be saved with data and a picture of the
strip concerning a data patient.

3.1 Start Microgel,Interlab G26, Pretty or ScanLab

Before starting Microgel, Interlab G26, Pretty or ScanLab software it is


necessary to open the archive and then the working database.

 Select the icon for Microgel or the command Start Microgel


from the menu.

 For Interlab G26, select the icon or the command Start


Interlab G26 from the menu.

 For Pretty, select the icon or the command Run PrettySoft.

 For ScanLab, select the icon or the command Run ScanLab.

Each time the menu command Start Microgel is selected, the Microgel
application will start automatically showing its the main window:

The menu command Start Microgel will be enabled only if Microgel is not
doing any analysis.

Each time the menu command Start Interlab G26 (Series 33) is selected, the

3.2 Data analysis

Once the analysis is completed and data have been transferred from the
Instrument, they will be automatically acquired by Elfolab where they can be
shown on the screen, analyzed and printed. During the data transfer the following
window will appear:

At the end of the transfer it will be possible to see the analyzed data.

3.3 Retrieve data

If it is needed to recalculate data already received using different calculus


parameters settings (this operation can be done in Calculus Setup), simply click
on menu Retrieve Data:

Select the desired Gels or sessions and push , at the end


of data transfer it will be possible to see the new graphs analyzed with the new
parameters.
If all traces should be recalculated, select the box “Select all”. In order to

exit from this window push the button.

3.4 Immunofixations (not for Pretty – ScanLab)

The Immunofixation is a method without reading that needs a scanner


to acquire the images of the analyzed Gels.
In order to acquire the immunofixation analysis results through the Scanner,
previously installed in the system, proceed as following:
1. From the Elfolab main panel click on menu Scanning IFE.

2. Insert the scanner sheet with the front placed toward the user.
3. Insert suitably the Gels with the front toward the scanner inside the frames in
the middle of the sheet.
4. In Elfolab select which Holders are going to be scanned: 1-2 or 3-4 for
Microgel and 1-4 or 5-8 for Interlab G26.

Click on the button to start the scanning of the Gels of the first two Holders

(or the first four for Interlab G26) and on for the Gels of the Holder 3 and 4
(or the last four for Interlab G26).

5. At the end of the scan it is possible to view the whole page with the button

(Full screen preview).

6. With the button it is possible to save the images in the selected folder
and in the database. It is possible to verify the settings going back to the main
window and checking that the images belonging to each patient are all shown
correctly.

With the button it is possible to select the d scanner:

With the menu “Immunofixations Crop Setup” it is possible to center the


image scanned or manage the width and the height. This menu has to be used only
if necessary.

When the scan is completed, it is possible to insert in the bottom of the


Elfolab main window, the Nephelometric values of the patient, if available. In
order to save the modifications it needs to click on button.
If the Sampling it was executed by the Interlab G26 instrument, it will be
possible to visualize the Dilution values and the Antisera names.
If the Sampling of Antisera was executed by the Interlab G26 instrument
(by the EasyFix method) the names of Antisera will be imported from the Interlab
G26 instrument.
It is still possible to rename the Antisera names, for the single
Immunofixation or for all Immunofixations.

If “Current Antiserum Name” is changed only the current trace will have the name

3.5 The ASD52 Sampler

The ASD 52 sampler is a very useful instrument for the electrophoresis


because it can automatically arrange up to 52 samples at the same time, read
the barcode of each test tube and send them to Elfolab software in order to
easily manage the patient’s personal data.
The bar code data can be received only if the Com Port of the PC
connected to the sampler ASD52 has been properly selected. This selection can
be done from the setup menu with the submenu of ASD52 Com Port. The
selected port will remain into the memory until a new port selection is done.

Each time the user wants to receive the bar codes, while the daily archive is open,

he can select the icon or the menu of the figure below.

After it has been selected, the Gel Holder to be associated to the bar code data
must be selected. After performing the selection and pushing “Ok” it will be
displayed a mask that will show the status of the bar code transfer.

When the Gel Holder or the session has been selected, press “OK” to read all the
bar codes. The following window will show the bar code reading result

According to the reading results it is possible to transfer all the received


IDs, just few of them or exit without importing any ID.
The window will show the present IDs and those acquired by the sampler.
It will also highlight those not reliable, non associated or repeated.
It will be possible then to import all the Ids, only those associated, only
those associated and not repeated or exit without importing any ID.

The Service ASD52 mask, protected by a password and for service use
only, can be utilized to receive, show, modify, print and save the Sampler
parameters.
In order to enter this window, select the menu “Settings” and then “Service
ASD52”.

3.6 Setup Chars Id

Elfolab allows selecting the characters received by the instrument Interlab


G26 with the Sampler (or by the ASD52) with the following menu.

In this case the following window will show up:

In this example the second received character will not be taken into consideration,
therefore if the read code is “1234” Elfolab will receive just “134”.
It is still possible receive only the values from “0” to “9”, without any chars.
You have to select the check “Transfer only the digits”. In this case, if the read
code is “A1234B56C”, Elfolab will receive just “123456”.

4 Software settings
The Settings menu allows a complete personalization of the software.

4.1 Language

Selecting Language on the main toolbar, will open another window


displaying a list of languages for selection. This choice can be modified at any
moment without any hitches for the work in progress. For example, it is possible
to print a file in each of the different available languages with no need of closing
and opening Elfolab.

4.2 Laboratory

Selecting Settings > Laboratory will open a window that allows editing
the information concerning the laboratory where the analyses are carried out. This
information is also printed on the patient file. The six Free fields can be used to

insert the titles of new fields to be utilized for personal data or hospital codes.

If the box “Repeat” beside the Free field is checked and the working archive
is open, the inserted value will be repeated for all the patient of the strip
(Microtech), Gel (Microgel) or Session (Interlab G26)

4.3 Choice of font

Selecting Settings > Font will display a window that allows choosing a
font, among those listed, for printing the file report. Central-European (CE) and
Greek fonts are also available.

4.4 Printing Type

Selecting Settings > Printing Type allows setting the printing mode of the
files. The changes done in this panel are saved in the memory

In this case the graph is not printed out but just fractions’ names and values.

The Chapter 5 will describe how to print the working databases data in a single
sheet still with the graphs.

4.5 Print Setup

Selecting Settings > Print Setup allows the selection of the fields to be included
in the printout for the 4 available factory pre-set printing types described above
(cfr paragraph 4.5).

If the yes checkbox is checked, the relative field will be printed.


It is possible, for example, not to include in the print the Interlab header, the gel or
trace number, the Free fields, etc…

It is also possible to select for each method the number of decimals (from 0 to 2)
for the analyzed traces.
The user can edit the Header of “Notes” and “Reviewed by ” field in the printout
and even decide at which point the “Reviewed by” field will be located.

In the left figure the “Responsabile” field starts at point 1100 of the horizontal
axis (and this is also the Default value), in the right figure at point 3500.
At last, the field “Axis Y Print Start” decides at which height should the printing
start for A4 format. In this way it is possible to create enough room for the
Laboratory headers. In the example below the printout starts at Y=2000
(Default=0)

4.6 Species Management

It is possible to manage up to 20 different species for animals and/or


controls.
Selecting Settings > Species Setup the user can “Create” , “Rename” and
“Erase” one species.

When the user selects “Create Species” he has to assign a name and press
OK.

When the user selects “Rename Species” he has to choose the Species to be
renamed, modify the name and press OK.

When the user selects “Delete Species” he has to choose the species to be
deleted and press OK.

It is also possible to select which specie will appear in the data transfer and
which will be the default one. In this last case press “Default Species” e select the
desired specie.

When Elfolab will receive data it will associate all record to the Default
Species. The user should eventually modify the desired Species directly from the
main mask.

For each created Species the user should setup the relative Normal Value
Range (cfr paragraph4.8)

4.7 Setting Normal Value Ranges

Selecting Settings > Normal Data the user can define the normal range of
values of each fraction, according to the type of analysis and Species.

setup: fraction names (up to 11 max) and their relative normal value ranges
expressed in % and, wherever allowed in the method, in concentration.
The Normal Values are present only for those methods that need reading.

main window.

In this way whenever Elfolab receive data from the Instruments, it will
automatically assign names and evaluate the analyzed fractions according to the
order setup in this mask.
It is suggested to modify names and default ranges soon after the
Software installation in order to work always with the same fractions from
the beginning.

4.8 Setup Calculus

Selecting Setting-> Setup calculus the calculus parameters are controlled


and setup.
It is clear that any change in this window will cause a different result for the
analysis, therefore it is strongly suggested not to work on them unless completely
sure regarding of the modification to be done.
It is suggested not to change these parameters without Interlab s.r.l.
approval.
Each method will have its own preset parameters.

The Calculus Setup is present only for those methods that require reading.

The parameters can change among the different methods. Some methods can have
less or more parameters with respect to others. Values of the same parameter can
differ among different methods too.
A description of the parameters follows below.

1. Start and End trace parameters


These parameters can isolate the graph part to show on screen. They decide
the Starting minimum point and the Ending maximum point of the analyzed
graph.
2. Delta Left, Shift Left, Delta Right
 DeltaSX: 1500 is the default value. This means that the first fraction is
singled out when the height of the curve, read from left to right of the
graph window, is equal to or greater than 1500.
 ShiftSX: 0 is the default value. In order to obtain a correct reading of
the graph, it is necessary to indicate how many points, before the x
point corresponding to the y value of 1500, the reading of Fraction 1
must start.
 DeltaDX: 200 is the default value. Same as explained for DeltaSX, but
in this case is referred to the last fraction. Therefore, the reading in the
graph window will be from right to left
3. Filter
This parameter filters received and calculated data, averaging every quote of
the graph with a number of previous and following points equal to the value
shown in the box.
4. Sensitivity for minima calculus
This indicates how many points of resolution the curve in the graph will
increase or decrease in order to obtain the recognition of a minimum. The lower
is the Sensitivity, the higher will be the number of minima.

5. Minimum percentage fraction

This shows the percentage value of the fraction area below which the area
itself is ignored in fraction recognition elaboration. The default value is set at
0.2%. In case a fraction has less than 0.1% it will not be considered as a fraction.
6. Accuracy
The accuracy parameter is used to adapt the value to a standard (as for the
control serum). Shifting this value will cause an increase or decrease of the
fraction value and proportionally all other fractions.
Example:
Accuracy is set to 7 for Fraction 1;
the software calculates the Fraction 1 equal to 63%;
on the screen it will be shown the value 63 + 7% (of 63) = 63 + 0.07*63 =
63 + 4.41= 67.41%.
The remaining fractions will decrease globally by 4.41% proportionally to
their value. The sum of all fractions will always be 100%.
7. Baseline
If the Baseline box has a check mark in it, it indicates that the baseline
will start from the minimum point detected within the graph. If the check mark is
removed the baseline will start from the minimum point detected on any point
along the full reading. The baseline is the Line on which the graph is placed.
8. Unit measures
The Unit measure box decides how to display the protein concentration in
grams/deciliter (g/dl) or grams/liter (g/l). For Urines, CSF and
Immunofixations the possibility are 3: milligrams/deciliter (mg/dl),
grams/deciliter (g/dl), grams/liter (g/l).

9. Choice of Micro or Semi-Micro Technique


Choosing the Semi-Micro technique will allow the analysis of patterns that
have a wider front on the bands, that are longer and therefore with a longer
scanning length. This results in the drawback of having a decrease in the number
of samples that can be analyzed per working cycle. Namely, when using the Semi-

micro technique on instruments like Microtech 648 4 samples per strip will be
produced instead of 8.

10. HbA2 Calibration


It is a numeric coefficient for the optimization of the HbA2 fraction value.
11. Recalc Min
If checked, it optimizes the minima calculus and makes the parameters
Filter, Delta and Edge, active too; with these it is possible to retrieve the
minimum position operating to the curve’s slope between minimum and the
previous maximum and between minimum and the following maximum.
12. Delta
The Delta parameter is active if Recalc Min is checked. It shows, in
percentage, the quote between a maximum and a minimum; the routine for new
calculus of minima is applied starting from this quote.
13. Edge
The Edge parameter is active if Recalc Min is checked. If minimum has a
value higher than the Edge, then no optimization is applied in its round.

14. Separation 27/14 and Separation 14/1


These parameters are present only for methods with 39 samples and these
show the OD points where the 3 traces of every strip are separated.

4.9 Color trace Intensity

This menu allows the modification of the color intensity of trace in the
right of graph.
If the number chosen will be high the color will be more intense.
The default values are set to 1.

It is necessary to highlight that Intensity values too high can help identifying
some bands but can also saturate high color values thus worsening the image
display.

These parameters are shown below.

1. Automatic Archive Backup


With this parameter enabled the software will automatically backup the
archive each time new data will be present.
For data security reasons, generally, this parameter must be enabled
in order not to lose data in case the PC suddenly turns off (i.e. in case of
power spikes).

2. Automatic saving of working database

If this parameter is enabled, each time the user moves from the daily
archive to the historical one, analyzed data will be automatically transferred to
the historical archive.
Otherwise at the archive closure the following question will be proposed.

In this case the user will decide to transfer or not data from the daily
archive to the historical one.
This function should generally be disabled in order not to lose data.
3. Gel ID input request at the daily archive closure
This parameter is useful for users that insert the Gel number in the “Gel
ID” field. In this case the parameter must be disabled.
When this parameter is enabled during the daily archive closure the following
question will be shown:

In order to remind the user that it will not be possible to insert the Gel Id in
the historic archive if it is not already present in the daily archive.

4. Analysis date modification on historical archive


This parameter can enable or disable the analysis date modification in the
historic archive.
This function is usually disabled.

5. Fraction Values

This parameter controls how to show the fraction values in the main
window. These values can be expressed in % or in concentration.
In the example below the parameter has been set to “Conc.”
It is always possible to show the values in % clicking on the “%” button.

6. Automatic Re-Assignment fractions names


This parameter is used for the Serumproteins method.
If it is enabled, whenever a minimum is inserted or removed (cfr.
paragraphs 6.2.1 and 6.2.2), the software will immediately calculate the name of
the fraction by checking if there are more or less than 5 fractions.
Otherwise the software will not recalculate the fractions’ names but will
simply remove the last fraction or add a new fraction.
Let’s clarify the concept.
In the example below the software has calculated 5 fractions upon data
receiving.

The user adds a new minimum and save the change.

If the automatic assignment is enabled (figure above) the software will add
a new fraction and rename all the old ones, inserting therefore Beta 1 and Beta 2
(6 fractions are now present).
Otherwise the software will simply add a new fraction leaving the old
fractions names unchanged.

This function must be enabled.


The only reason to disable it is to maintain special fractions names
already assigned that the user doesn’t want to recalculate.

7. Enable Data filter


If this parameter is enabled, received data will be filtered from the reading
noise.
This function should generally be enabled.

8. Account Management

In order to use the Account Management Select Yes in the window for Account
Management. Select Save then exit from Elfolab program and re-open.

Change Password

To modify the password, select Settings > Change Password:

The following window will appear:

Insert a password and repeat. An error message will appear if the password is not
repeated correctly.

Account Levels

There are 6 levels that can be set up within the software for allowing users to
access specified areas.
In order to define the levels the Administrator can go in Settings > Setup Account
Levels.

Account Management

Once each level is defined, the administrator can create, name and specify
the passwords for each account in Settings > Account Management.

In order to remove an user the Administrator must select the name in the list
and push the button Canc.

It is possible to change operator at any time selecting the menu .

9. Criterion List Import from Host


This Parameter allows selecting how matching the patient’s data imported from an
Host PC (see paragraph 1.4).

 In the “Matching Sample ID” mode the data imported from Host Pc will
be matched in according to the order of “Sample Ids” already present on

Elfolab. The “Sample IDs” present in the Host List, but not in Elfolab, will
not imported in Elfolab.
 In the “Sequential” mode the data imported from the Host PC will be
inserted in Elfolab in according to the order of Host List.

In the “Sequential” mode it needs to insert the tubes in the


instrument exactly in according to the order of Host List.

10. Sessions Management (only for Interlab G26 series 36, 42 and 48)
With the Session Management the customer can work in two modes:
 Automatic: The instrument G26, when the Analysis is started,
scans the barcode of the method and the Id gel directly from the gel
and then it transmits them to Elfolab. Elfolab become available in
the working database the number of records required for the
method recognized by the instrument. The customer no longer
has to select the method to each session, to avoid possible
mistakes caused by manual selection.
 Manual: The user chooses the method for each session. The G26
instrument will not scan the bar code of the gel. It is the standard
method of working.
This Setting is available only before to open the database.

11. Save Patients Without Scanning


With this parameter, the user can decide to save in the historical database
patients data that do not require reading as, in example, immunofixations. In
such way the user can trace any analysis and eventually print some comments
on the analysis printout.
This function is usually disabled.

12. Validation
This parameter allows the user to select which result data transmits to the

LIS. If Yes is selected here, the reviewer must select the icon in the main

panel. Upon results validation, the icon changes to and the result will
export to the LIS.

13. View Graph


This window allows selecting different colors for the graph display in the
main window and in the printout as well as for the text inside the graph.

14. Select Import-Export Files

This Panel allows to select the files name for Import and/or Export files (see
page 29).

2. In the mask select the Import (or Export) directory and the file name
(alternatively it possible to create a new file writing the new name in the
field “File name”).
3. Click “Open”.

the IFE images will be exported during the data export.

15. Elfolab Master

The user can decide to work with a database connected in a central


location. The user can select the button in order to select the
database.
This feature allows multiple systems/instruments to share a common
database.

The possible mode in order to use Elfolab Master with 2 (or more) Elfolab
is the following:

 in one of two PC with Elfolab create a shared folder;


 from one Elfolab make a Backup of database (see page 111) and
insert this copy in the shared folder;
 for each Elfolab go in Settings->General Parameters and with the
button select the Backup copy in the shared folder;
 for each Elfolab click “Yes” on ElfoMaster Parameter.

4.11 Setting Host

This window displays the parameters available for Host connection with
shared files or with serial Port.

f Memorize the Import Directory is set to No, each time the patient
demographics are requested to upload to Elfolab, the system will require the
selection of the import directory.

5 Databases Management

The Archive, or Database, is a list of patient data. The Archive is divided in


a historical section and daily section. Data received from the Instruments are first
loaded in the daily archive; then it is possible to definitively store them in the
Historical or General section which will therefore continuously increase.

5.1 Opening a Database

Once the program is started, the user has to open the database from Menu
Database -> Open Database.

The archive is opened as “historical” (Cfr. Chapter 1 A new day of work,

pg.24). When the user enters the temporary database, by clicking on , it is


then possible to insert data from new patients. These new patients’ data can then

be stored in the historical database by clicking on or by choosing the


Save last reading option in the Database menu.
After saving the last reading, the database will include the new patients data.

If the user exits Elfolab without saving the last reading, it will be still be
possible, even after days, to use the data (doing corrections, completing patients’
data entry etc.) by retrieving the archive. This option is active provided that, after

clicking on for patient data entry, the following window is displayed

and press “Current Working Database”. This choice will in fact keep the
previously acquired data in the temporary database. If the button “New daily
Archive” is selected instead, the temporary database records will be reset, namely
canceled, in order to allow the insertion of a new set of data (cfr. Chapter 1).

5.2 Patient data Entry

This operation can be accomplished by (cfr. 1.3 Inserting Patient


pg.27):

5.3 Viewing the List of Patients

This option is available in the File menu and allows the user viewing the
complete list of the patients in the current database and eventually to print is (cfr.
Chapter 1.5).
After inserting the patients personal data or at any time during this process,

it is possible to click on in the File menu to view the Patient List with all
the relevant data about them.

5.4 Navigating in the Database

The exploration of an archive can be easily accomplished by using the keys


shown on the bar at the bottom of the Elfolab main panel.

After the selection have been made, on the main window the patient data

graphs contained on the selected gel, session or strip:

The window shows the graphs along with fractions names and values,
highlighting in red those who are out of the normal range.
With the button at the bottom of this window, the user can move to the next
gels (or sessions or strips) to view their traces.
The print button starts the printout of this window.

5.5 Inserting Notes

During the personal data insertion and after having reviewed analysis
results, it is possible to add notes to the patient record.
Three options are available:

a) Clicking on ;
b) Using the Archive menu;

c) Clicking on the Notes field in the main window ;

The Patient Note mask can perform different functions but it is also very
easy to use.

The first of these is writing a specific note for an individual patient. Click in the
green area and begin inserting text. Click the Save button to record the note.

The patient notes screen is divided into two areas: the NOTES OF PATIENT and
the ARCHIVE OF NOTES.

NOTES OF PATIENT

This area contains the notes of the current patient. The note doesn’t have any length
limit.

The following three options can be used in the Notes of Patient


 Edit Note.
 Delete Note.

This area can store an unlimited number of notes that can be used for all patients.

Confirm. If it is in Archive of Notes, first select the note through the scroll-bar

5.6 Patients Search

With this version of Elfolab 4 different filter options are available for the
patient search. This makes much easier to reconstruct the electrophoretic history
of a patient.

The 4 different search options are described below:

Filter: The Archive can be browsed with the help of a filter that can
be applied on the different data fields of the database. A filter is the

union of all characteristic that the patients must have in order to appear in the list. Patients who
don’t have those characteristics will not be cancelled from the database but simply hided until the
filter is disabled.

5.6.1 Data Filter

This is the classic version of the filter.


It can be used only when the Historical database is open.
In order to open the historical database, click on Patient History under the
Database menu.

The following window will appear in which the user can put data in the
various fields.

When all the conditions have been inserted, click on “OK” button and only
the filtered patients will be shown in the main window. Results viewing, print and
delete operation will be done only on these filtered patients.
On the message bar at the bottom of the window the number of filtered
patients, among all present in the Archive, corresponding to the filter conditions
will be shown.

The filter can be removed clicking on button or choosing Remove


filter from the Archive menu.

5.6.2 Patient’s History

This new and very important function, working both in the Historical and
Daily Archives, shows in real time whether the current patient is already present
in the Historical database.

If the patient is already present the button will be enabled and


colored, otherwise it will be disabled and gray.
The user can setup this automatic search using the menu Setting->Filtered
Patient Setup

In the following window it is possible to select the fields to be used for the
automatic search.

The Default field is the Name, but user can choose one or more additional
fields from the available ones.
If more than one field is selected, the current patient will be identified
as already present only if there is a patient in the database having all the
fields coincident with the selected ones.

If the patient is already present click on button to enter the Patient’s


History mask.

All available records for the filtered patient will be shown in the grid at the
bottom of the window. The Current patient will receive number 1.

Double click in a different row of the grid to compare current patient data
with past data. This row will receive number 2.

Two overlapping graphs will be displayed. It is possible to print the two


graphs either in A5 format (one above the other) or overlapped.

Click the button to view all images found in a grid.

The button can connect permanently the two data previously


overlapped. In this way the software will save the link between the two results
(cfr. paragraph 5.6.4).
The link can be removed with the button “Remove link”.

5.6.3 Automatic Filter

The button enabled the automatic filter according to the criteria


setup of the Setting menu->Filtered Patient Filter

The automatic filter avoids entering the filter window (cfr. paragraph
5.6.1).
This button is enabled only in the Historical Archive.

5.6.4 Data Link

When two records have been linked (cfr. paragraph 5.6.2) the software
keeps it in memory forever. In the main window, when a linked record is

selected, Elfolab enables the button. After clicking on this button Elfolab
will filter all the records linked to this current one. This function is quite
important when, in example, the user wants to recall the immunofixation linked to
serumproteins data of a patient. The records must be linked first as described in

paragraph 6.5.2 and then the link is recalled with the button.

In order to remove the filter click .


This button is enabled only in the Historical Archive.

5.7 Deleting Patients

User can delete a patient file only if he is working with the historical
archive.

Select the patient to be deleted and show his data on the screen, then use the
Delete Patient function from the Database menu:

The elimination of a patient file is irreversible. This command can also be


applied either to a set of Patients that have been selected by the Filter option (cfr.
paragraph 5.6) or to all the Patients of an archive.
Choose the desired option and click “GO!”.

In order to avoid that the user delete data by mistake it is necessary to


protect the program with a password.

5.8 Pathologic Database

If a patient is considered pathologic it is possible to store it in a dedicated

sub-database, simply check the box . The patient will be


automatically saved in the sub-database.
To search for a pathologic patient previously saved perform a data filtering
(paragraph 5.6.1) and select the pathologic check box.

5.9 Moving from Working Database to Historical Database

During working work it is possible to consult the historical database.

Push the button to open it.

In order to return to the daily session push and select “Current Daily
Archive” in the following window:

5.10 Backup Copy of Database

Elfolab can perform, each time it starts, one or more Backup Copies of the
working database.
This operation is very important and should be done on regular basis to save
the working data. If the PC fails and no backup has been done on a CDROM or a
Pen Drive, it may happen that all data would be lost.

To make a backup copy of database, before opening the working database,


select the menu “Database”-> “Run Backup Database”.

The Backup database will be named Backup.mdb but and the user can
modify its name and the destination folder.

This menu is enabled only before opening the database.

It is important to have some backup copies and to save them in a


CD/DVD or Pen Drive. In this way the customer won’t lose data if some
problem occurs to the PC.

5.11 Browsing Old Archives

Through the menu Database->View an old database, Elfolab can show,


filter and print data from old Elfolab database versions or Backup copies of the
current working database.

The user can select the desired “.mdb” file from the following window.

This menu is enabled only before opening the database.

Obviously, when an old archive is opened, data modification commands


are not allowed and the working archive cannot be opened at the same time
because these archives cannot be used anymore to receive new analysis data.

5.12 Compact Database

When the Elfolab database reaches the maximum size, Elfolab will
automatically compact the database and remove old data from the database in use.
These data will be reallocated in a backup database in the installation folder of
Elfolab. The database will named with the date of the backup and it will can be
used for consultation.
From the menu Settings>Settings Compact Database it is possible to set
the number and the type of records that it will be removed at any compaction.

It will be possible to remove all the old data, only the data not pathological,
only the data without note or only the data not pathological and without note.
We recommend to use the setting “Not pathological and Without note”; in
this case the pathological data and the data more important will remain in the
main database.

Elfolab provides a wide number of functions that allows you to operate in
the graph, including corrections and measurements.
All available functions are shown as icons in the panel of the main window
just underneath the graph.

Some methods may have some commands disabled.

6.1 Commands Description

6.1.1 Adding a minimum

The icon allows for the addition of a minimum to a chosen point in the
graph layout.
Click on the icon to activate the function. A plus symbol will appear near
the mouse pointer. Move the pointer to the position where you want to add the
minimum and click. A yellow vertical segment will appear to indicate that the
minimum was fixed. All fractions will be automatically recalculated.

Adding a minimum is one of the most used functions. The


software is already setup to add a minimum and therefore
Note! simply click with the left button on the desired point to perform
the command.

6.1.2 Deleting of a minimum

The icon allows for the deletion of any minimum from the graph.
Click on the icon to activate the function. A minus symbol will appear near
the mouse pointer. Move the pointer to the position where you want to delete the
minimum and click. The minimum, shown by the yellow vertical segment will
disappear.

Deleting a minimum is one of the most used functions. The


software is already setup to remove a minimum and therefore

6.1.3 Establishing a new baseline

The icon allows you to change the baseline of the graph. Clicking on the
icon and keeping the left button of the mouse pressed a horizontal white line will
appear.

Move the pointer to the position where you want to establish the new
baseline and release the mouse button to fix the baseline. Moving the pointer to
the left or to the right will make the baseline tilt.

Baseline: is a line that allows excluding from the calculation the part of the
Note! graph under the baseline itself.

6.1.4 Measurement of the percentage area

The button allows you to measure the percentage area of a part of the
graph.
Click on the icon to activate the function then move the pointer to the part of
the graph that you want to measure. Choose the starting point, then keep the left
button of the mouse pressed and move the pointer. You will see the percentage
value displayed on the graph this will vary according to the movement of the
pointer. Move the pointer until the area of interest has been completely measured
and then release the button. This function may prove to be useful when trying to
have a rough estimate of some monoclonal components on the pattern.

6.1.5 Modifications of the graph shape

Both and buttons allow you to modify the graph shape by drawing
segments and rectangles respectively.

Click on the icon(s) to activate the function(s). Choose the starting point on
the part of the graph to modify and then, keeping the left button of the mouse
pressed, move the pointer to the final position. Releasing the button will display
the effect of the selected icon on the graph.

6.1.6 Establishing a new point of origin on the graph

Click on the icon to activate the function. Move the mouse pointer on
the graph, a yellow vertical line will appear. Keeping the left button of the mouse
pressed, move the mouse pointer to the new point of origin on the graph (any
position on the graph).

6.1.7 Shifting the graph

Click on the button to activate the function. Move the mouse pointer on
the graph a red vertical line will appear near the mouse pointer. Keeping the left
button of the mouse pressed, move the mouse pointer to the left or to the right.
You will see the graph shift to the left or to the right according to the position of
the red vertical line.
Releasing the mouse button will result in a modified position of the graph.

6.1.8 Graph Ruler

In all the methods except Serum Proteins and LDH isoenzymes, it is


possible to use the button to display a ruler at the lower part of the graph.
Clicking on the icon will display a ruler that indicates the points (expressed
as pixel) that are read along the entire length of the gel holder.
The ruler allows you to set the start and end parameter values that are useful
for the Setup Calculus of Hemoglobin and Lipoprotein methods (cfr. 4.7 Setting
Normal ).

6.1.9 Optical Density

Clicking on the button will open a window that allows you to view the
reading values recorded by the instrument.
This option allows you to see what has been really read by the instrument,
without any elaboration, namely the rough data.

shifting the blue vertical line along the graph, by moving the mouse pointer.

6.1.10 Optical Density –Zoom Effect

The user can enlarge any part of the graph area by moving the mouse
pointer to the selected starting position; then, keeping the left button of the mouse
pressed, move the pointer until you reach the end point.

Releasing the left button of the mouse will result in the magnification of the
selected area of the graph.
To restore the initial display, click on the right button of the mouse.

6.1.11 Optical Density – Edit Cut Graph

In the Optical Density Panel, for Methods Serumproteins 5 or 6 fractions for


39 samples it is possible to modify the Start and End for single traces. This
possibility is available only in the working database and only for the last Session
data received. In the historical database it is not possible to modify the data from
the OD Panel.
This tool can be useful for example if in the trace is present an extra gamma
band. In this case the software automatically can not cut correctly the graph.
In the following example is present a case extra gamma band.

In this case it is possible:


 Click on the “OD” button in the Main Panel;
 Select from Menu “Edit Cut Traces” the related Cut;
 Select from the Red (or Green) Cursor the Separation Point;
 Accept the Change;
 Exit from the OD Panel;
 Confirm the Change;

6.1.12 Start and End of traces

Two functions have been added for all methods for correction in the graph
window, except Serumproteins and LDH Isoenzymes. To activate them click on
one of the following two buttons:

6.1.13 Fraction recalculation

The button is used to correctly recalculate the position and values of


the fractions in case any minimum has been manually added or deleted (cfr.
paragraphs 6.1.1 and 6.1.2).

6.1.14 Monoclonal Components

If the analysis performed is a Serum Protein, with the use of button, it


is possible to select up to 8 suspected monoclonal components (SMC).
For the use of this function see Chapter 8.

6.1.15 Reference Trace

The commands are used to setup and show a Reference Graph.

With the command the current graph is taken as Reference Graph.

With the button a comparison between the current graph and the
reference one is shown.

6.1.16 Quality Control

The button is used to insert the current data into a Quality Control (cfr.
Chapter 7).

6.1.17 Graph Save

The command saves the current graph in text format, with extension
“.txt”.

The command saves the current graph in image format, extension


“.bmp”.

6.1.18 Cancel Modifications

The command cancels all the modification operations done on a graph


and restores the original one.

The command cancels just the last modification performed on the graph.

6.1.19 Filter on the graph

The icon is used to smooth a part of the graph. Choose the starting point
on the graph and, while keeping the left button of the mouse pressed, move the
pointer to the final position.

The intensity of smoothing depends on the Filter value in the setup calculus
section. It is possible for one or more smoothing events to take place in the same
area.

6.1.20 Cancel the graph

The Command allows to cancel a graph. The command can be utilized


if the graph is not scanned correctly. In this case the patient won’t be saved in the
historical database.

7 Quality Control
7.1 Quality Control

With Elfolab is possible to calculate quality control for both precision and
accuracy of results.
The steps to perform the quality control are the following:

a) Choose the traces that will be considered useful for the C.V. calculation
(Precision);
b) Create a database where the data will be inserted;
c) Put the data into the database that was just created;
d) Control the values: Average, Variance, Standard Deviation, CV and
Levey-Jennings curves.

Following are the steps in detail.

7.2 Entering into the Q.C. windows

In order to enter into the Q.C. windows: from the temporary archive
(database), select method and gel or strip to analyze and start the analysis.
When data have been received from the Instrument and the graph correction

has been eventually completed, click on the button located in the Elfolab
main window of the graph correction panel (cfr. paragraph 6.1.15).
If the daily archive is active and additional data need to be inserted all at the
same time, it is possible to access the QC even from the Menu Quality Control-
>Insert data in QC

At this point the following message will appear:

If the answer is Yes, the data will be added to the same QC file as the last
one entered. In this case if the current trace is not compatible with the Method of
last QC a message will appear:

If the answer is “Yes” data will be added to the last calculated QC


otherwise answer “No” to add data in a new or different QC archive

7.3 Initialize a New QC

When the answer to the following question is “New QC” it will be required
to insert the base information before creating the new Archive.

Required data are QC name and method and it is also possible to change the
normal value ranges of the QC. Such range by default will be the same as the
Normal Value, but the user at this step can change it to his desire.

When the first record is inserted in the QC it will not be


possible to modify the Name, Method and normal value range

If the working archive is open, the window will show the desired gel(s)/session(s)
and traces. In this way all the desired records of the working archive could be
inserted in a single moment.

When the selection has been made, click on the “Insert” button.
If a QC with the same name is already present a message will appear and the
user will have to change the name:

7.4 Data Display and Print

When one or more data have been inserted, Elfolab will show is a table all
the records of the QC together with the basic information for each present
fraction: Average, Variance, Standard Deviation and CV.

just the main data of the QC for all the fractions as shown in the following figure.

considered not reliable.

In this case the whole grid will become red and the user has to click on the
row to be deleted.
The panel below opens the Levey-Jennings curves for each present fraction.

In this mask the Levey-Jennings curve is shown together with the Standard
Deviation and Normal Value intervals.

Through the Panel it is possible to select


the elements to be shown and then printed.
From this window it is possible to print the QC values ( “PrintQC” button)
of the current fraction or the graph with the Levey-Jennings curve ( “Print Chart”
button).

8 Automatic monoclonal components

8.1 Automatic SMC

It is now possible, for the Serumproteins method, to find automatically the


presence of one or more monoclonal components in the electrophoretic trace.

For methods different from serumproteins a similar search


procedure exists and is able to highlight interesting areas.
Note!

The software automatically will find the most correct monoclonal zone
inside the selected area.
In this way the error of the manual assignment, variable from user to user,
will be eliminated.
The usage of this function is pretty easy.
Whenever a monoclonal band is detected in the gel or in the graph with the
symbol it is sufficient to select the button at the left hand side of the graph in the

main window .

Then the window for the SMC selection opens.

8.2 SMC window

The SMC selection window looks like the following figure:

The graph shows the same trace image of the Elfolab main window but
bigger and more defined.
All the functions of this window are explained in detail in the following
sections, anyway they are very intuitive and easy to use.

8.2.1 Automatic SMC selection

In order to start this function, simply press the button. The


function will remain enabled each time the SMC window is opened.

In order to select the SMC it is needed to select the start and end of the
area using the left button of the mouse. As soon as the start position is selected, a
blue line will be shown in that point.

To remove the start point already chosen, press the right button of the
mouse.
Once the end position has been selected too, the software will analyze the
area and, in case the SMC is found in that zone, will fill the whole area with a
color.

Additional data will show the SMC value as a percentage of the fraction
where it has been found, as a percentage of the total graph and, pushing

button, it will be possible to see the absolute reading value,


normalized to the maximum value of the graph and the eventually value in
concentration.
The absolute value is very important because, if a modification is done on
part of the trace that doesn’t include the SMC, the percentage values of the
SMC will surely change, while the absolute value of the SMC area will
remain unchanged.
If the software doesn’t find any SMC in the selected area it will show the
following message:

To remove the SMC select the button at the left hand side of the values.
To hide the SMC (which will remain in the memory) simply click on

button.

8.2.2 Manual SMC selection

It is possible to choose the SMC start and end manually, as it happens in


the graph correction window.

In such case the button to select is . In this case the user will
directly choose with the mouse the start and end position of the SMC, without any
search by the software.
All the SMC information, described in paragraph 8.2.1. will be shown
anyway.

8.2.3 Fraction visualization

By selecting the menu View->Fractions, the frame with the fraction values will be
displayed (by default already present at the mask opening). If the fractions colors
are different from the default, the relative areas will be filled with the same color
as well.
To remove the frame simply select again the button.

8.2.4 Print

Trough the button it is possible print in A4 format the graph in


which the SMC has been found.

To show the meaningful minimums present in the trace simply select the
menu View->Minimums.
To remove the minimums select again the same button.

To show the meaningful maximums present in the trace simply select the

To show the meaningful flexes present in the trace simply select the menu
View->Flexes.
The flexes colors will be different, purple or blue respectively if the flex is
upward or downward.

8.2.8 Filter

The filter is needed to remove the noise caused by a non optimal data
transfer. The noise can cause problems to the automatic SMC search.
In these cases it is possible to filter the curve in order to remove the noise
before searching for the SMC.
In order to do so, select the “Filter” button and choose with the left mouse
button the start and end point of the area to be filtered. When the start point has
been selected, a blue line will appear showing the chosen position.
In the following figures it is possible to see the gamma zone before and after
the filter. A correct SMC will be found only after having performed the filter. The
curve will change back to the original.

8.3 Anomalies Alarm

By default the software is setup to highlight the presences of graph


anomalies in the gamma zone (cfr. paragraph 8.4), in other words the presence of
a non typical shape of this zone. Not always these anomalies indicate the presence
of one or more SMC but the software notifies anyway that the trace could require
additional analysis.

When this happens, the software shows the signal in the Elfolab main
window.

Enter the SMC window and select the button to identify the irregular
areas in yellow color.
To hide the “Spike” areas just lick again the same button.

8.4 Setup windows

In the SMC window there are 3 Setup menus that can be used to modify

some settings .

8.4.1 Parameters Setup

In the parameters window some essential settings of the automatic SMC


search can be modified as well as the list of Serumproteins fractions that turn on
the SMC Alarm notification (cfr. 8.3 ). By default the warning is enabled only for
the gamma zone.
The main parameters are password protected because only the Interlab
technical support can access and modify them.
The user must not modify the parameters in the window below unless
under direct guidance from Interlab support.

8.4.2 Print Setup

Through this window, it is possible to select the SMC values that will be
printed on the report.

If the Total value is printed, then it is not possible to print the % Fraction
value regarding the corresponding fraction and vice versa.

If the SMC concentration is printed, then it is not possible to print the


Absolute Value regarding the corresponding fraction and vice versa.

To save or cancel the changes press the relative buttons.

8.4.3 Color Setup

In the color setup window it is possible to select both the fractions colors
and that of the eventual SMCs present in the SMC window.
By default the colors are the following:

By default the fraction have neutral color.

To modify any color simply click in the colored box that has to be
changed:

Then the following color window appears and the color can be chosen.

At the end of the modifications the changes can be saved pushing the
button, otherwise press to cancel or to restore the default
colors.
