collides with the active site of its enzyme, an enzyme-substrate complex forms (Figure
5.5b). As a result of binding to the enzyme, some of the chemical bonds in the substrate are
weakened. The substrate then undergoes chemical change, the product or products are
formed, and the enzyme detaches.
Enzymes generally have a high degree of specilcity; they catalyze only one type of
reaction, and most act on only one particular substrate. An enzyme’s shape (its tertiary
structure; Chapter 2, p. 42), especially the shape and electrical charges at its active site,
accounts for its specificity. When an enzyme acts on more than one substrate, it usually acts
on substrates with the same functional group or the same kind of chemical bond. For
example, proteolytic, or protein-splitting, enzymes act on different proteins but always act on
the peptide bonds in those proteins.
Enzymes are usually named by adding the suffixase to the name of the substrate on
which they act. For example, phosphatases act on phosphates, sucrase breaks down the sugar
sucrose, lipases break down lipids, and peptidases break peptide bonds. Enzymes can be
divided into two categories on the basis of where they act. Endoenzymes, or intracellular
enzymes, act within the cell that produced them. Exoenzymes, including extracellular
enzymes, are synthesized in a cell but cross the cell membrane to act in the periplasmic space
or in the cell’s immediate environment.

Properties of Coenzymes and Cofactors


Many enzymes can catalyze a reaction only if substances called coenzymes, or cofactors, are
present. Such enzymes consist of a protein portion, called the apoenzyme, that must combine
with a nonprotein coenzyme or cofactor to form an active holoenzyme (Figure5.6).

A coenzyme is a nonprotein organic molecule bound to or loosely asso-ciated with an


enzyme. Many coenzymes are synthesized from vitamins, which are essential nutrients
precisely because they are required to make coenzymes. For example,coenzyme A is made
from the vitamin pantothenic acid, and NAD (nicotinamide adenine dinucleotide) is made
from the vitamin niacin. A cofactor is usually an inorganic ion, such as magnesium, zinc, or
manganese. Cofactors often improve the fit of an enzyme with its substrate, and their
presence can be essential in allowing the reaction to proceed.
Carrier molecules such as cytochromes and coenzymes carry hydrogen atoms or
electrons in many oxidative reactions (Figure 5.7). When a coenzyme receives hydrogen
atoms or electrons, it is reduced; when it releases them, it is oxidized. The coenzyme FAD

biasanya bekerja pada substrat dengan gugus fungsi yang sama atau jenis ikatan kimia yang
sama. Sebagai contoh, proteolitik, atau pemecahan protein, enzim bekerja pada protein yang
berbeda tetapi selalu bertindak pada ikatan peptida pada protein tersebut.

Enzim biasanya dinamai dengan menambahkan sufiksasi ke nama substrat di mana


mereka bertindak. Sebagai contoh, fosfatase bertindak pada fosfat, sukrase memecah gula
sukrosa, lipase memecah lipid, dan peptidase memecahkan ikatan peptida. Enzim dapat
dibagi menjadi dua kategori berdasarkan di mana mereka bertindak. Endoenzyme, atau enzim
intraseluler, bertindak di dalam sel yang menghasilkan mereka. Exoenzyme, termasuk enzim
ekstraseluler, disintesis dalam sel tetapi melintasi membran sel untuk bertindak di ruang
periplasmik atau di lingkungan terdekat sel.

Sifat-sifat koenzim dan kofactor

Banyak enzim dapat mengkatalisasi reaksi hanya jika zat yang disebut koenzim, atau ada
kofaktor. Enzim tersebut terdiri dari bagian protein, yang disebut apoenzim, yang harus
bergabung dengan koenzim atau kofaktor nonprotein untuk membentuk holoenzim aktif

Koenzyme adalah molekul organik nonprotein yang terikat dengan atau secara longgar terkait
dengan enzim. Banyak koenzim disintesis dari vitamin, yang merupakan nutrisi penting
justru karena mereka diminta untuk membuat koenzim. Misalnya, koenzim A terbuat dari
asam pantotenat vitamin, dan NAD (nikotinamida adenin dinukleotida) terbuat dari vitamin
niacin. Kofaktor biasanya berupa ion anorganik, seperti magnesium, seng, atau mangan.
Kofaktor sering meningkatkan kecocokan enzim dengan substratnya, dan keberadaannya
dapat menjadi penting dalam memungkinkan reaksi untuk melanjutkan.

Molekul pembawa seperti sitokrom dan koenzim membawa atom hidrogen atau
elektron dalam banyak reaksi oksidatif (Gambar 5.7). Ketika koenzim menerima atom
