Activity Of Amylase :

Collection of 10 Drops
of Saliva in a big test tube.

Add

3 Drops of starch solution to the

prepared saliva in the test tube

Add

3 drops of IodineAdd
solution and stir

"solution should be light blue"

 Record the time of formation of blue color

Let the solution stand for few minutes and

record when the blue color disappears

Transfer

5 drop of solution from step #4

to the test tube Transfer

Add

2 ml of Fehling A&B.

 Place in water bath for 2-3 minutes.

Activity of Catalase

A) Extraction of catalase

Peel and shred the potato with the grater.

Soak with 50mL of water

Filter through cheesecloth

Place 5ml of Filtrate into test tube

Add

5ml OF 6%H2O2 up to 3 cm
mark in test tube.

Insert a glowing wooden

spint

Observe the formation of gas by

* The action of the potato enzyme on the hydrogen peroxide

 Measure the height of the foam after 1 minute .

Measure from the top of the liquid to the top of the foam.
 Measure the foam height every minute for a total of 5 minutes.
 Construct a graph of foam height on the y-axis versus time on the x-axis.
Label as graph 1
Repeat B step #1 with the following test tube.

Place each tube at a constant temperature

for several minutes.

- Test tube #2 – ice bath (0c)

- Test tube #3 – room temperature bath (20-25C)
- Test tube #4– body temperature bath (37-40C)
- Test tube #5–hight temperature bath (70-80 C)

Remove the tubes from the baths

ADD

H2o2 up to 3 cm in the test tube

*Put the tube back into the bath.

 Compare the rate of Gus production :

by measuring the foam height in each tube for 5 minutes , similar to step #5.
 Construct graph, similar to step #1 , for each test tube

B) Effect of PH :

Place potato pulp in 3 test tube to

depth of about 2cm.

label the test tubes 1,2 and 3

  To test tube #2, Add 5 drops of 10% NAOH.
 To test tube #3, Add 5 drops of 10% HCL

Determine and record the Ph of solution in the three test tube.

- Add H2O2 up o 3cm Mark of the test tube .

- Measure the foam height in each tube after 1 minute
