Efek Penyembuhan Luka Bakar dari Ekstrak Buah Mengkudu (Morinda

Citrifolia L.) dalam Sediaan Gel pada Kelinci

Abstrak

Mengkudu (Morinda citrifolia L.) merupakan salah satu tumbuhan

bermanfaat. Buahnya dapat digunakan sebagai obat luka bakar. Tujuan dari
penelitian ini adalah untuk memformulasikan ekstrak buah mengkudu dalam
bentuk sediaan gel yang baik dan mengetahui efek penyembuhan luka bakar
dari sediaan gel.
Ekstrak buah mengkudu diperoleh melalui perkolasi dengan etanol 70%
dan diuapkan dengan penguap putar untuk memperoleh ekstrak kental. Gel
dibuat dari natrium karboksilmetilselulosa dan diformulasi dengan ekstrak
buah mengkudu. Kadar ekstrak buah mengkudu dalam sediaan gel terdiri atas
1, 3, 5, 7, 9, 11, 13, 15%. Sediaan dievaluasi, yaitu pemeriksaan organoleptis,
homogenitas dan pH selama 28 hari. Sediaan gel diuji terhadap luka bakar
dengan mengoleskan sediaan secara merata pada permukaan luka. Luka bakar
dibuat dengan lempeng besi bulat panas bersuhu 80oC dan berdiameter 2,1 cm
dengan kelompok kontrol yaitu kontrol yang diberikan formula basis gel,
tanpa pengobatan, dan Bioplacenton®. Sediaan gel ekstrak buah mengkudu
yang memberikan efek penyembuhan paling cepat kemudian dibandingkan
dengan sediaan gel di pasaran (Bioplacenton®). Pengujian dilakukan pada
kelinci putih jantan. Pengamatan dilakukan secara visual dengan mengamati
perubahan diameter luka setiap hari. Analisis data dengan Uji T Sampel Bebas
menggunakan Statistical Program Service Solution (SPSS).
Hasil penelitian menunjukkan ekstrak buah mengkudu dapat
diformulasikan dalam sediaan gel dan tidak terjadi perubahan organoleptis,
homogenitas dan pH dari sediaan gel ekstrak buah mengkudu selama 28 hari
dan nilai pH yang diperoleh adalah 5,1 sampai 5,8. Kelompok yang diberikan
gel yang mengandung ekstrak buah mengkudu 1, 3, 5, 7, 9, 11, 13 dan 15%
berturut-turut sembuh pada hari ke-19, 18, 13, 15, 15, 15, 15, 17 dan diperoleh
hasil gel ekstrak buah mengkudu 5% memberikan efek penyembuhan paling
cepat yaitu pada hari ke-13. Kelompok kontrol yang diberi basis gel tanpa
ekstrak sembuh pada hari ke-26, sedangkan kelompok yang tanpa pengobatan
sembuh pada hari ke-31 dan kelompok yang diberi Bioplacenton® sembuh pada
hari ke-10. Hasil yang diperoleh bahwa Bioplacenton® memberikan efek
penyembuhan lebih cepat dari gel ekstrak buah mengkudu 5%, tetapi tidak
berbeda signifikan secara statistika. Dapat disimpulkan bahwa gel ekstrak buah
mengkudu dapat digunakan untuk menyembuhkan luka bakar.

 Effect of Burn Healing of Indian Mulberry Fruit Extracts (Morinda
citrifolia L.) in Gel Formulations toward Rabbit

Abstract

Indian mulberry (Morinda citrifolia L.) is one of the efficacious herbs.

The fruit can be used as a burn medication. The aims for this research was to
formulate the indian mulberry fruit extract into a good gel dosage form and to
know the effect for burn healing from gel dosage form.
The indian mulberry fruit extract was obtained by percolation used 70%
ethanol and evaporated by rotary evaporator to obtain the condensed extract.
Gel was made from carboxylmethylcellulose sodium and formulated with
indian mulberry fruit extract. The concentrations of indian mulberry fruit
extracts in gel formulations were 1, 3, 5, 7, 9, 11, 13, 15%. The formulations
were evaluated, that were organoleptic, homogenity, and pH test for 28 days.
Gel dosage form tested against burns by dabbing dosage forms smoothly on the
surface of the wound. The burn wound done by round iron hot plate
temperature of 80o C with diameter 2,1 cm and as the control groups were
control used gel base formulation, untreatment group, and Bioplacenton®. Gel
of indian mulberry fruit extract that gave the most rapid healing effect then
compared with commercial gel dosage form (Bioplacenton®). The examination
was treated to white male rabit. Observations made visually by observing
changes in diameter of wound every day. Analyzed data with Independent
Sample T Test used Statistical Program Service Solution (SPSS).
The results showed the indian mulberry fruit extracts could be
formulated into gel dosage form and there were no organoleptic, homogenity
and pH changes of indian mulberry extract gel formulations for 28 days and the
pH values obtained was 5,1 to 5,8. The group was given gel that contained
indian mulberry fruit extract 1, 3, 5, 7, 9, 11, 13 and 15% respectively
recovered by day 19, 18, 13, 15, 15, 15, 15, 17 and obtained result that gel of
5% indian mulberry fruit extract gave the most rapid healing effect by day 13.
Control group that given gel base without extract recovered by day 26, whereas
the untreatment group recovered by day 31 and group that given Bioplacenton®
recovered by day 10. The result was obtained that Bioplacenton® gave more
rapid healing effect than gel of 5% indian mulberry fruit extract, but there was
no significant difference statistically. It can be concluded that gel of indian
mulberry fruit extract can be used to heal burn.
