Abstrak
Iodin merupakan trace elemen (elemen substansial) yang harus tersedia dalam tubuh untuk
pembentukan hormon tiroksin. Namun, besarnya asupan iodin yang tidak normal (kelebihan
atau kekurangan) dapat menimbulkan masalah kesehatan, khususnya pada kelenjar tiroid
(hipotiroid ata hipertiroid). Jumlah iodium yang disekresikan melalui urin dapat
menggambarkan jumlah iodium yang dikonsumsi. Oleh karena itu, pemeriksaan kadar iodin
dalam urin dirasa perlu untuk dilakukan guna mendeteksi gangguan tiroid. Metode yang
digunakan dalam pemeriksaan kadar iodin urin dalam praktikum ini adalah APDM
(Ammonium Persulfate Digestion Method), di mana sampel di-digesti dengan ammonium
persulfat. Reaksi yang berlaku adalah reaksi Sandell-Kolthoff, di mana iodida yang
terkandung dalam urin berperan sebagai katalis saat As3+ mereduksi Ce4+ menjadi Ce3+. Hasil
reduksi Ce4+ menjadi Ce3+ menimbulkan perubahan warna yang terdeteksi oleh microplate
reader, yang kemudian diinterpretasikan sebagai kadar iodin. Pembacaan absorbansi Iodin
dilakukan pada panjang gelombang 405 nm. Kurva baku menghasilkan persamaan garis y =
1,1707x - 6,7677 dengan nilai r2 0,9762. Hasil perhitungan kadar iodin pada sampel urin 1-5
secara berurutan yaitu 5,475; 5,528; 5,416; 5,430 dan 5,401 ppm. Terdapat beberapa faktor
yang dapat memengaruhi hasil analisis, di antaranya jarak waktu pengambilan sampe urin
dengan waktu pengukuran, jarak waktu penambahan reagen, serta teknik dalam
menggunakan micropipet.
Abstract
Iodine is a trace element (a substantial element) that must be available in the body for the
formation of thyroxine hormones. However, the amount of abnormal iodine intake (excess or
deficiency) can cause health problems, especially in the thyroid gland (hypothyroid ata
hyperthyroidism). The amount of iodine secreted through the urine may reflect the amount of
iodine consumed. Therefore, examination of urinary iodine levels is considered necessary to
detect thyroid disorders. The method used in examining urinary iodine levels in this
practicum is APDM (Ammonium Persulfate Digestion Method), in which the sample is
digested with ammonium persulfate. The reaction based on Sandell-Kolthoff reaction, which
the iodide contained in the urine acts as a catalyst when As3 + reduces Ce4 + to Ce3 +. The
reduction of Ce4 + to Ce3 + results in a color change detected by the microplate reader, which
is then interpreted as iodine content. Iodine absorbance readings were performed at a
wavelength of 405 nm. The raw curve yields the equation of the line y = 1,1707x - 6,7677
with the value of r2 0.9762. The results of calculation of iodine content in the urine samples
1-5 sequentially ie 5.475; 5,528; 5,416; 5,430 and 5,401 ppm. There are several factors that
can affect the results of the analysis, including distance time of urine sampel taking with
measurement, the time of addition of reagents, and techniques in using micropipet.
