ISOLATION OF ACTIVE ANTIOXIDANT COMPOUNDS FROM EXTRACT ETIL

13161048 Background of the study: The plants of Simpur Philippine (Dillenia philippinensis Rolfe) include the Dilleniaceae family which has benefits as antimicrobial, antihyperglycemic and antioxidant. In the previous study it was found that Simpur Philippine contain triterpenoid compounds, flavonoids and phenols. Leaf ethyl acetate extract has a very strong antioxidant activity. Purpose of the Study: This study aims to isolate the antioxidant active compound of ethyl acetate extract of the leaf Simpur Philippine (Dillenia philippinensis Rolfe). Methods of the Study: Extraction was performed by a multilevel reflux method using n- hexane solvent, ethyl acetate and methanol. Fractionation and purification using vacuum liquid chromatography and classical column chromatography. The obtained isolates were tested for purity using thin layer chromatography with three single and two dimensional developers. Then, the pure isolate was characterized and identified using UV-Vis spectrophotometry and IR spectrophotometry. Antioxidant activity test using free radical damping DPPH method. Result of the Study: DP isolate had antioxidant activity which reacted to give yellow color with DPPH 0.2% spotting appearance. Identification of DP isolates with shear reagents has a maximum wavelength of 364 nm and 264 nm which is a flavonoid compound and allegedly a class of flavonols. From the IR spectral data the DP isolates contain functional groups of O-H, C = C aromatics, C-H alkyl, C = O, C-H, and C-O. Conclusion : DP isolate had anticrosidan activity with 32% inhibition at 70 μg / mL concentration and was thought to be flavonoid flavonoids with OH substitution on atoms C 3.5.6 and 7.
