BAGIAN HISTOLOGI
Learning Outcome

The student will be:

 Explained about bone marrow histology
 Explained about component and function of vascular niche
 Explained about component and function of endosteal niche
 Explained about blood cell tranlocation from bone marrow to sinusoid
lumen.
 Red bone marrow  erythroid progenies
 Yellow bone marrow  adipose cell
 Interchangeable
 Connsist of:
 Stromal
 Hematopetic cords
 Sinusoidal capillaries
 Hematopoetic Stem Cell
 Committed precursor cell
 Maturing cell
 Vascular niche
 Endosteal niche
Vascular niche
 Blood vessels surrounded by non hematopoetic stromal cells
 Mesenchymal stem cells
 Adipose cells
 Provide a local source of energy
 Synthesize growth factors
 The population of adipose cell increases with age, obesity and following chemotherapy

 Endothelial cells
 Phagocytic activity
 Produce growth factors that stimulate the proliferation and differentiation of hematopoetic cells.

 Reticular stromal cells

 Produce hematopoetic growth factors and cytokines that regulate the production and differentiation
of blood cells

 Macrophages
 Remove apoptotic cells, residual nyclei from orthochromatic erythroblast and megakaryocytes
 Exclude partcles from entering the marrow
Endosteal niche

 Located at the endosteum-bone marrow interface

 Consist of:
 Preosteoblas
 Osteoblast:
 Produce multiple hematopetic cytokines: G-CSF, M-CSF, GM-CSF, IL-1, IL-6, IL-7

 osteoclast
RBC

 Biconcave shape for high surface/volume ratio

 Main function is oxygen and carbondioxide transport
 Contain haemoglobin
 Have no cell organelles
 Cell membrane is braced by an actin/spectrin-containing cytoskeleton
which maintains shape
eosinophil

 Have bilobed nucleus

 Eosinophilic granules
 Ganular contain of eosinophil:
 Eosinophil peroksidase
 Binds to microorganism and facilitates their killing by macrophages

 Major basic protein

 Binds to and disrupts the membrane of parasites
 Causes basophils to release histamine
neutrophils

 Main role : phagocytosis and bacterial killing

 Neutrophils cytoplasma contain three types of granules:
 Primary :
 Similar to lysosomes
 Contain acid hidrolases
 Contain myeloperoxidase
 Secondary
 Specific to neutrophil  involved ini the mobilization of inflammatory mediators and
complement activation
 Tertiary
 Contain enzymes, may be involved in the phagocytic process
basophil

 Bashophilic granules contain :

 proteoglycan heparin
 chondroitin sulphat
 histamine
 leukotriene 3
 Have highly specific membrane receptor of IgE
