 Medication is distributed through out the body ,given
by oral ingestion or parenteral.
 For most therapeutic agents only a small proportion
of the medication reaches the site of action.
 Eg ; in chemotherapy of cancer – 99% of the drug do
not reach to the targeted tumour site.
Targeted drug delivery system
 Targeted to specific parts of the body where there is
solely diseased tissue is present , thereby avoiding the
interaction with healthy tissue.
 The goal of the T.D.D.S is to prolong, localize , target
and have a protected drug interaction with the diseased
tissue.
Advantages of targeted drug delivery
system:

 Reduce the frequency of dosage taken by patient


 Having more uniform effect of the drug
 Reduction of drug side effects.
 Reduces the interaction of drug with healthy tissue
and prevents the damage.
 Helps in the stem cell therapy of myocardium tissue.
Types of targeting methods
 Passive targeting: achieved by cloaking the nano
particles with some sort of coating with substances
like PEG .
 Results in the antiphagosytic substance (hydrophobic),
thus it is able to circulate in the body for longer
duration

 Active targeting: drug loaded nanoparticles enhances


the targeting
 nanoparticles could posses the capability to be
activated by a trigger that is specific to the target site ,
such as using ph responsive material .
NANO SPONGE
WHAT IS NANO SPONGE?
 Nano sponges are tiny sponges with a size of
virus(250nm -1µm)
 Which consist of cavities that can be filled with a wide
variety of drugs.
 The sponge acts as a three dimensional network or
scaffold which consist of the backbone as long length
polyester.
 Nano sponges are tiny mesh like structures made up of
naturally degradable polyester.
 These tiny sponges can circulate around the body until
they encounter the specific target site and stick on the
surface and begin to release the drug .
PREPARATION OF NANO
SPONGES
 Nano sponges are prepared by polymers and cross
linkers.
 One of the method for preparing nano sponge is
reacting cyclodextrin with a cross linker such as di-
isocianates, diaryl carbonates.
 The average diameter of formed nano sponge is below
1µm but fractions below 500 nm.
ADVANTAGES OF NANO
SPONGES:
 A wide variety of drugs can be loaded into nano sponges,
carries both lipophilic and hydrophilic substances and
can improve the solubility of poorly water soluble
substances.
 Nano sponges are solid in nature and can be formulated
as oral, parenteral, topical or inhalational dosage forms.
 Can be used to mask unpleasant odours and flavours.
 Hydrophobic substances can be encapsulated as they
are water soluble.
 Nano sponge formulations are stable over range of pH 1
to 11, and at temperature up to 130˚c.

DISADVANTAGES:
 Only small molecules can be inserted.
 Dose dumping may be take place.
 May retard the release.
