5A. Details of professional training and research experience, specifying period.

I have a considerable amount of research experience starting with my M.Sc


dissertation work. I have acquired skills and expertise in the areas of microbial
fermentation, enzyme purification and crystallisation, biochemical characterisation of
proteins, and enzyme-mediated reactions in aqueous and non-aqueous media.

I have worked both in academic and industrial environments and made significant
contributions in India, the United Kingdom and Germany.

B. Details of employment

a) Worked as a Post-doctoral Scientist at the Robert Koch Institute, Nordufer


20, D-13353 Berlin, Germany (January - December 2003).
Nature of duties included: the one-year project entitled "Biochemical
characterisation of the phospholipases A of amoebae as natural hosts of the
bacterium Legionella" involved work on the following 5 objectives:

1. Investigation of the secretion and intracellular accumulation kinetics of


phospholipase A-activity (-ies) of/in Acanthamoeba castellanii and its
(their) substrate specificity.
2. Partial biochemical protein purification of the phospholipase A-activity
(-ies) of A.castellanii culture supernatants/cell lysates, classification of
the activities according to their substrate specificity, and approximate
determination of their molecular weight.
3. Determination of phospholipase A-activity compared to uninfected
amoebae at several time points Acanthamoeba castellanii infection
with Legionella pneumophila.
4. Investigation of intracellular multiplication of Legionella pneumophila
in Balamuthia mandrillaris.
5. Analysis of Balamuthia mandrillaris culture supernatants/cell lysates
for phospholipase A-activity.

b) Worked as Advanced Development Scientist in the Inhalation Drug


Delivery division of 3M Healthcare Limited, Ashby Road site, 3M House,
Morley Street, Loughborough, UK (July – December 2000).

Nature of duties included: biochemical work on deoxyribonuclease I


(DNase I), an enzyme for the treatment of cystic fibrosis in human beings;
development/validation of assay methods; purification and
characterization; formulation of the enzyme in metered dose inhalers and
studies on the stability of the formulations.

c) Worked in a supervisory role in the Quality Assurance department of a


sandwich-filling-producing food production plant, M/s. FreshPak Chilled
Foods Limited, Kernan Drive, Belton Industrial Estate, Loughborough,
UK (February – May 2000).

Nature of duties included: Supervision of the maintenance of


microbiologically sterile conditions in the production plant, new product
development; and a member of the taste panel of the company.

d) Worked as a post-doctoral fellow in a project on the purification,


crystallisation and structure-solution studies of a phospholipase from the
venom of Red Scorpion under the supervision of Professor T.P.Singh at the
Department of Biophysics, All India Institute of Medical Sciences, New
Delhi (August – November 1999).

Nature of duties included: biochemical characterisation of the


phospholipase; purification of the enzyme; crystallisation; and testing of
the anti-microbial activities of some natural and synthetic peptides.
