Share DBT & CSIR SPONSORED National Workshop on Plant-Biotechnology RAPD-PCR techniques On August, 18th 20th, 2011at ANNAMALAI UNIVERSITY. HOST DEPARTMENT The Department of Botany has an Outstanding Record ofpromoting Excellent Research, right from its inception. It has been recognized as DST-FIST & UGC-SAP Sponsored Department. The Department offers 2 year M.Sc. Botany and 5 year integrated M.Sc. Herbal Science Programme. In addition, M. Phil. and Ph.D. programs are offered. The Department has produced 161 Ph.Ds in a variety of botanical disciplines such as Biodiversity and its Conservation, Genetics, Cell and Molecular Biology, Microorganisms and Plant Pathology, Pollution and Environmental impacts on Ecosystem, Plant Physiology and Biotechnology, Plant Taxonomy and Plant Genomics. The Department has organized workshops, seminars and refresher courses through which a huge number of participants from various universities and institutes took part and have placed benefits. WORKSHOP BACKGROUND The Polymerase Chain Reaction (PCR) is a technique that permits geometric and enzymatic amplification of specific sequences of DNA. The PCR is now widely used in research laboratories and doctors offices, relies on the ability of DNA-coping enzymes to remain stable at high temperatures. No problem for Thermus aquaticus, the sultry bacterium from yelllowstone that now helps scientist to produce million of copies of a single DNA segment in matter of hours. In nature, most organisms copy their DNA in the same way. The PCR mimics this process, only in test tubes. When any cell divides, enzymes called polymerase made a copy of the entire DNA in each chromosome. Besides, based on the applications, PCR has been modified in a number of ways to suit specific application situation, several molecular markers (RFLP, AFLP, ISSR, SSR, SNP and RAPD) also available for analyzing genetic variations at molecular level. Among these, RAPD is an inexpensive and a rapid method not requiring much information regarding the genome and has been widely used to document the genetic diversity in plants and animals. Marker-based genome analyses have become increasingly important for various fields of plant science research. In this background, the present workshop will help to provide researchers a hands-on training in molecular marker techniques and to highlight the importance of marker assisted techniques in the plant biotechnology research. PROGRAMME SCHEDULE Morning Session : 9.00 am to 1.00 pm;

Lunch : 1.00 pm to 2.00 pm Afternoon Session : 2.00 pm to 6.30 pm Tea break in the middle of each session (15min.) WORKSHOP PROGRAMME Besides hands-on training, participants will be encouraged to carry out the techniques themselves. The workshop will be conducted for 30 participants only. This workshop of RAPD-PCR technique will cover guest lectures and hands-on experience on various aspects of biotechnology as follows Laboratory Safety Tools- Knowledge-information regarding chemicals and other basic instruments required Major instruments PCR-thermal cycler, Electrophoresis, Gel documentation. Chemicals preparation for DNA isolation and purification. DNA quantification Agarose-gel Electrophoresis PCR reactions Techniques to be handled DNA extraction: Isolation and purification of genomic DNA (plant sample). DNA quantification in spectrophotometer : DNA qualitative analysis in agarose-gel electrophoresis Amplification in PCR thermal cycler Initial denaturation, Denaturation, Annealing and Extension. Agarose-gel Electrophoresis, Gel-documentation, Data scoring,Data analysis in software Results and interpretation REGISTRATION The participants are requested to fill-in the Registration form and mail it to the Organizing Secretary along with a demand draft for Rs. 2000/- from Indian Bank / ICICI Bank / SBI in favour of The Registrar, Annamalai University payable at Annamalainagar or Chidambaram.
