Stockage Intended use For in vitro diagnostic use. Polyclonal Rabbit Anti-Guinea Pig Immunoglobulins/HRP, code No. P 0141, is intended for use in immunocytochemistry (1), immunoblotting and ELISA (2). P 0141 may be used either for the direct detection of guinea pig immunoglobulins, or as a secondary antibody for the detection of a guinea pig antibody (primary antibody) that has reacted with its target antigen. Reagent provided P 0141 is the purified immunoglobulin fraction of rabbit antiserum conjugated with horseradish peroxidase (HRP) of very high specific enzymatic activity. The coupling reaction is a modification, developed at DakoCytomation, of the two step glutaraldehyde method of Avrameas and Ternynck (3). The reaction is gentle, efficient, highly reproducible and gives conjugate molecules of Mr predominantly 200 000 to 240 000. The conjugate is provided in liquid form in 0.05 mol/L Tris/HCI, 15 mmol/L NaN3, pH 7.2. Immunoglobulin concentration g/L: See label on vial. Immunogen Specificity Immunoglobulins isolated from guinea pig serum. The antibody used for conjugation has not been absorbed to remove cross-reactions with immunoglobulins from other species and therefore shows a particularly strong reaction with guinea pig immunoglobulins of all classes. The antibody may cross-react with immunoglobulins from other species, for example mouse and rat. Precautions 1. For professional users. 2. This product contains sodium azide (NaN3), a chemical highly toxic in pure form. At product concentrations, though not classified as hazardous, sodium azide may react with lead and copper plumbing to form highly explosive build-ups of metal azides. Upon disposal, flush with large volumes of water to prevent metal azide build-up in plumbing. 3. As with any product derived from biological sources, proper handling procedures should be used. 4. The product may be used in different techniques and in combination with different sample types and materials, therefore each individual laboratory should validate the test system applied. P 0141 kann entweder fr den direkten Nachweis von Meerschweinchen-Immunglobulinen oder als sekundrer Antikrper fr den Nachweis eines Meerschweinchen-Antikrpers (primrer Antikrper) verwendet werden, der mit seinem Zielantigen reagiert hat. P 0141 ist die gereinigte Immunglobulin-Fraktion des Kaninchen-Antiserums, konjugiert mit Meerrettich-Peroxidase (HRP) sehr hoher spezifischer enzymatischer Aktivitt. Bei der Kopplungsreaktion handelt es sich um eine von DakoCytomation entwickelte Modifikation der zwei Schritte umfassenden Glutaraldehyd-Methode von Avrameas und Ternynck (3). Die Reaktion ist schonend, effizient wie auch hochgradig reproduzierbar und erbringt Konjugatmolekle mit einer relativen Moleklmasse berwiegend zwischen 200 000 und 240 000. Bei den in diesem Produkt verwendeten Konzentrationen kann Natriumazid, obwohl nicht als gefhrlich klassifiziert, mit in Wasserleitungen vorhandenem Blei oder Kupfer reagieren und zur Bildung von hochexplosiven MetallazidAnreicherungen fhren.
