3 ml reagen 0,3 ml serum

CARA KERJA
Vortex

diamkan selama 1 menit

Baca absorbansi spektrofotometer (=340 nm)

Hidupkan stopwatch

Baca absorbansi lagi setelah 1 menit, 2 menit, dan 3 menit

ASAT activity (U/L)= A/min sample x 1745

DISKUSI
Apa yang anda ketahui tentang AST?
Apakah fungsi AST bagi metabolisme didalam
tubuh?
Mengapa AST bukan indikator yang spesifik bagi
Liver?
REAGENT COMPOSITION
R1:1. TRIS (pH=7,65) 110 mmol/L
2. L-Aspartate 320 mmol/L
3. Malate Dehydrogenase (MDH) 800 U/L
4. Lactate Dehydrogenase (LDH) 1200 U/L
R2:1. 2-Oxoglutarate 65 mmol/L
2. NADH 1 mmol/L
TEST PRINCIPLE
2-Oxoglutarate L-Aspartate

AST/GOT

L-Glutamate Oxaloacetate NADH+ H+

MDH

L-Malate NAD+

NADH is oxidized to NAD+, the resulting decrease in absorbance at 340

nm is directly proportional to the activity of GOT in the sample.
This is a modified formulation for the assay of GOT, as recommended by
the IFCC (International Federation of Clinical Chemistry).

Aspartate Transaminase
(SGOT)
AST is called serum glutamic
oxaloacetic transaminase (SGOT).
Aspartate aminotransferase (AST, EC
2.6.1.1) is enzymes found mainly in
the liver, but also found in red blood
cells, heart cells, muscle tissue and
other organs, such as the pancreas
and kidneys.
1 = Malate dehydrogenase
2 = Aspartate aminotransferase
Aspartate Transaminase (AST), also known as serum
glutamic oxaloacetic transaminase (GOT) or aspartate
aminotransferase (ASAT/AAT), facilitates the conversion of
aspartate and -ketoglutarate to oxaloacetate and
glutamate.
There are two isoenzymes in humans:
GOT1 is a cytosolic isoenzyme derived from red blood cells and
heart;
GOT2 is the mitochondrial isoenzyme found mainly in the liver.
AST is elevated in liver and muscle diseases. It is part of
diagnostic tests for liver function, myocardial infarction,
acute pancreatitis, acute hemolytic anemia, severe burns,
acute renal
