In normal practice, quantities is carried out after design stage and before construction stage. Why after design stage ? Because to carry out taking off, detail drawings from design stage is needed. Lets see example :

The purpose of bill of quantities To set down the various items of works in a logical sequence so that it readily priced by contractor.
ii. Enables all the contractors to tender on the same information iii. Provide a basis for the valuation of variation if any changes involved. What is bill of quantities ? A list of items giving the quantities and brief description of work to be carried out with is respective quantity and unit cost for carrying out each work.
